Optic nerve injury is commonly involved in ocular trauma as well as glaucoma as a characteristic change. Recent investigations suggest that changes of micro-environment for retinal ganglion cells (RGC) and axonal regeneration may enhance survival and axonal regeneration. In the series of our experiments using fluro-gold retrolabelling technique, immunohistochemical staining, and animal model, we observed that Schwann cell-derived neutrophilic activity (SCNA) and ciliary neurotrophic factor (CNTF) could promote cultred retinal cells and survival of GRC and axonal regeneration in a rat model of cruched optic nerve injury. Those results provide basis for further study for clinical application.
视神经损伤是一种常见致盲眼外伤,尚无有效疗法。本项目拟提取和应用雪旺细胞源性营养神经活性物质及睫状神经营养因子,应用荧光金逆行标记、免疫组化染色、银染法等技术观察其对培养的视网膜节细胞及视神经非横断性损伤模型中节细胞的存活与轴突再生的作用,比较不同给红方法的效果,为深入认识视神经再生和节细胞拯救及临床应用提供依据。
