Necrotic enteritis (NE) is a worldwide poultry gastrointestinal disease caused by the overgrowth of Clostridium perfringens. It is necessary to fully understand inflammatory response mechanisms of chickens with NE for developing effective approach to control the disease. Study on NOD-like receptors(NLRs)/NF-κB signaling pathway may provide new ways for exploring the inflammatory response mechanisms of chickens with NE. The present study will build the necrotic enteritis (NE) model by infecting broilers with Clostridium perfringens to study the effects of NE on NLRs mRNA expressions on spleen and intestines of broilers. In vitro, primary chicken intestinal epithelial cells and spleen cells will be isolated to study the effects of Clostridium perfringens and its toxins on inflammatory responses of these cells through silencing NLRs/NF-κB signaling pathway genes for exploring whether NLRs/NF-κB signaling pathway was involved of activating inflammatory response of chickens with NE. In addition, further study will be carried out to investigate whether lactic acid bacteria will alleviate inflammatory responses of chickens with NE through regulating the NLRs/NF-κB signaling pathway.
产气荚膜梭菌感染引起的肉鸡坏死性肠炎(NE)是危害世界肉鸡生产的一种胃肠道疾病。全面了解NE肉鸡的炎症反应机制对发展有效的防控NE措施是必需的。NOD样受体(NLRs)/NF-κB信号通路的研究可为探讨NE肉鸡炎症反应的分子机理提供新的途径。本项目通过感染产气荚膜梭菌建立肉仔鸡NE模型,研究NE对NLRs在肉仔鸡脾脏和肠道中基因表达的影响,并通过在体外细胞培养体系中运用RNA干扰技术沉默NLRs/NF-κB信号通路上关键分子来研究产气荚膜梭菌及其毒素对肉仔鸡肠上皮细胞和脾脏细胞的炎性反应影响,以探索NLRs/NF-κB信号通路是否参与激活NE肉仔鸡的炎症反应。在此基础上,进一步研究日粮中添加乳酸菌是否可通过调控NLRs/NF-κB信号通路的关键分子从而缓解NE肉仔鸡的炎症反应。
