Sphincter of Oddi is a complex and special anatomic structure of smooth muscle groups with duodenal junction ,but independent of the duodenum outside. Its' structure and regulationg mechanism plays an important role in SO diastolic and systolic motion. When these structures altered, SO functional disorder will happen. As well-known, intracellular free Ca+2 in somooth muscle contraction plays an extremely important role as "second messenger". Recent study indicated that isolated SO has rhythmic contraction function in the weak foundation of tension based, and was regulated significantly in a dose-dependent of somatostatin in the microenvironment. The study also indicated this rhythmic contraction function have a certain relationship between the acute pancreatitis and somatostati. Present study will use cell patch-clamp technique to further explore the biological characteristics and regulating mechanism of SO motor by calcium channel, and to further investigate SO function state and its role in the process of AP, and relationgship between the two; Study of somatostatin on SO function in pancreatitis will be further discussed in order to find the relationgship between SO function state and acute pancreatitis prognosis, also,the effect and mechanism of somatostatin on SO in the treatment of AP process.
Oddi括约肌(sphincter of Oddi, SO)是包绕在胆管、胰管及其共同通道末端的极为精致且高度协调的一群与十二指肠连接,但又独立于十二指肠的平滑肌群,其结构及调控机制在SO运动功能方面发挥着重要的作用。当这些调节因素紊乱时可发生SO舒缩功能的异常。平滑肌细胞胞浆游离Ca2+在平滑肌收缩过程中起着极为重要的"第二信使"作用。在我们前期的体外研究中发现,正常离体SO在其微弱基础张力的基础上具有节律性的舒收功能,且与急性胰腺炎的发生以及生长抑素及其受体之间存在着一定的相互关系。我们将在此工作基础上应用细胞膜片钳技术研究SO平滑肌细胞膜上钙离子通道对SO舒缩功能的内在调节机制;并在此基础上探讨SO的功能状态在急性胰腺炎(AP)发生过程的作用,以及二者的相互关系,明确Ca+2通道、SO功能状态以及二者在AP转归过程中发挥的作用;同时探讨生长抑素在治疗AP过程中对SO功能的影响作用
