Based on the theory of Pharmaphylogeny, adhering to the theory of traditional Chinese medicine doctrine,as the starting point of the biological basis of correlation between the Chinese medicine closely related species Rhizoma Panacis Japonici and Rhizoma Panacis Majoris, to explore the law for the association between the genetic diversity of medicinal plants and the material potency of traditional Chinese medicine(TCM), to present a novel approach for TCM modern research on the basis of plant genetic relationship. The ISSR-PCR will be performed to mark the DNA of three plant species from different populations to evaluate the genetic diversity. In addition, the RNA of the plants will be sequenced thoroughly in order to compare the differences of functional gene structures and their expression levels. Then the type and content of saponin (common ingredient) will be analyzed by chemical means. Finally, pharmacological methods will be employed to analyze the correlation between the gene transcription levels and the clinical functions. Collectively, this study aims: to attempt to illustrate the phylogenetic relationship between Panax japonicus C.A.Mey and Panax japonicus C.A.Mey. var. major (Burk.) C. Y. Wu et K. M. Feng and Panax japonicus C.A.Mey. var. bipinnatifidus (Seem.) C. Y. Wu et K. M. Feng at the molecular level, to analyze the correlation between plant germplasm resources and quality of herbal materials, to explain the correlation of the phylogenetic relationship of plants, herbs, chemical composition, pharmacological effects and Indications of association between Rhizoma Panacis Japonici and Rhizoma Panacis Majoris, to interpret the substance of its scientific connotation, to provides a theoretical studying foundation and scientific basis for clinical connotation.
基于药用植物亲缘学的理论,秉承中医药的传统学说。本项目以研究中药近缘种之间关联性形成的生物学基础为切入点,以竹节参,珠子参药材为例,探索药用植物遗传多样性与中药材药性之间的关联规律,在此基础上提出基于植物亲缘关系的现代中药研究的思路与方法。 拟采用ISSR-PCR标记不同居群3种植物的DNA,评价其遗传多样性;进行植物RNA的深度测序,在RNA水平上比较植物重要功能基因结构和表达水平的差异规律;运用化学和药理的方法分析共性成分皂苷类型与含量、相关基因转录水平与主治功能的关联性。本研究首次尝试在分子水平上阐明竹节参和变种间的亲缘关系,分析药用植物种质资源与药材品质的相关性,阐释竹节参和珠子参2种中药材间的"植物亲缘关系-药材-化学成分-药理作用-功能主治的关联规律",解读其科学内涵的实质,为规范竹节参和珠子参临床用药提供了理论研究的基础和科学依据。
本项目基于药用植物亲缘学理论,以五加科人参属的竹节参及其近缘种为对象,研究竹节参居群之间及居群内部的遗传多样性和复杂性,结合分子生物学和生物信息学手段分析不同药材化学成分的含量、结构类型及其相关基因转录水平与药性的关联性。目前,我们通过该项目完成了全国范围内的竹节参资源调查,并首次进行了竹节参和珠子参的转录组测序工作,同时完成了竹节参和珠子参的指纹图谱的分析和对比工作。本研究成果将为阐明竹节参植物的进化特点和次生代谢调控机制提供理论基础和科学依据,为我国中医药研究领域提供了创新性的研究思路和方法。
