Esophageal carcinoma's early diagnosis and prognosis is very difficult and the patients’ treatment outcome is poor. The NS5ATP9 gene, whose protein can be combined with the proliferating cell nuclear antigen (PCNA), may be associated with the growth and development of many kinds of tumors. Our previous study observed that the NS5ATP9 gene is highly expressed in esophageal tissue, and related to patients’ overall survival and prognosis. Based on this conclusion, this project intends to further observe the miRNAs which may target the NS5ATP9 gene would be verified. Their role in esophageal carcinoma mechanism, their expression in the plasma or serum of esophageal carcinoma patients and their relationship with clinical diagnosis and prognosis would be clarified by the Bioinformatics technology, microRNA microarray, animal experiment techniques, etc and the clinical follow-up and case-control study. This project would provide useful clues for further understanding the function of NS5ATP9 gene and its related miRNAs, applying them as noninvasive clinical bio-marker for esophageal carcinoma.
食管癌的早期诊断和判断预后不易,其治疗效果较差。NS5ATP9基因表达的蛋白能够与增殖细胞核抗原(PCNA)结合,研究表明其与肿瘤的发生发展相关。我们前期发现NS5ATP9在食管癌细胞组织中高表达,且与食管癌患者的总生存期和预后密切相关。在此基础上,本课题拟进一步通过生物信息学技术联合肿瘤相关miRNAs芯片、动物成瘤实验等方法筛选并验证可能靶向作用于NS5ATP9基因的内源性miRNA分子,研究这些miRNAs在食管癌中的作用机制以及在食管癌患者血清血浆中的表达情况。最终通过临床随访与病例对照阐明NS5ATP9基因及其相关miRNAs与食管癌临床诊断和预后的关系,为进一步了解NS5ATP9基因的功能和将其应用于临床作为食管癌早期无创诊断标志物提供有益的线索。
食管癌的早期诊断和判断预后不易,其治疗效果较差。研究表明NS5ATP9基因与肿瘤的发生发展相关。本课题在前期发现NS5ATP9在食管癌细胞组织中高表达,且与食管癌患者的总生存期和预后密切相关的基础上,通过生物信息学的方法筛选出高度可能靶向作用于NS5ATP9 基因的内源性miRNA 分子(miR-216a-5p、miR-183-5p、miR-19a-3p、miR-19b-3p、miR-200b-3p、miR-200c-3p、miR-429),并应用荧光素酶报告基因法和miRNA-mRNA复合物分析法得出miR-216a-5p抑制NS5ATP9表达的结论。同时,通过观察类似物及抑制剂对细胞行为学的变化,发现miR-216a-5p抑制肿瘤细胞的增殖和迁移能力。并通过标本验证及临床随访分析证实了血浆miR-216a-5p的表达水平是食管癌患者总生存期及肿瘤复发的独立影响因素。另一方面,我们发现NS5ATP9基因在结直肠癌中也存在表达差异,并且其表达在放射照射后随时间增加。NS5ATP9高表达可以促进直肠癌细胞放疗照射后的细胞增殖性,同时调控肿瘤细胞周期。并且在结直肠癌患者的组织及外周血标本中,我们发现miR-183独立表达升高,可以作为潜在的预测肿瘤复发和预后的标志物。
