Based on the view of "Salty into kidney" from neijing, and combined with modern scientific research, the research want to propose the hypotheses that salt-sensitive hypertension is located in the kidney, and to enrich the theory of the etiology and pathogenesis of the Chinese salt-sensitive hypertension. The experiment will use the data mining method to discover a salt-sensitive hypertension in ancient Chinese medicine, to further verify the theoretical hypothesis of disease in the kidney. The experiment will present scientific hypotheses from the kidney-specific molecular expression to explore the mechanism of salt-sensitive hypertension molecules, combined with salt-sensitive hypertension genomics research.The kidney-specific cadherin expression experiment will study in-depth to explore the expression of abnormal mechanisms leading to salt-sensitive hypertension, which will propose a new molecular mechanism of salt-sensitive hypertension, but also will expand the scientific content of the theory of "Salty into kidney" from the traditional Chinese medicine.
根据《内经》"咸入肾"理论,结合现代科学研究,提出盐敏感性高血压病位在肾的理论假说,丰富了中医盐敏感性高血压的病因病机理论;采用数据挖掘方法对中医古籍进行盐敏感高血压相关内容的发掘,进一步验证病位在肾的理论假说;结合目前盐敏感性高血压的最新基因学研究成果,提出了从肾脏特异性的分子表达中探寻盐敏感性高血压分子机制的科学假说;为了初步验证上述假说,又以肾特异性钙粘素的表达为研究对象,并深入探讨其表达异常导致盐敏感性高血压的机制,从而提出了盐敏感性高血压的新的分子机制,同时也扩大了中医"咸入肾"理论的科学内涵。
本项目包含文献数据挖掘和动物实验两个部分。文献数据挖掘部分通过对中医古代文献的研究,整理出了高血压医案数据库,并采用数据挖掘技术对其进行了研究。动物实验部分采用Dahl所建立的DS大鼠,随机分为三组:模型组、模型+药物组、空白组。模型组大鼠五周龄投以8%食盐饲料;模型+药物组投以8%食盐饲料同时给予中医补肾方药提前干预;空白组投以普通饮食。观测相关指标。结果发现:1、通过中医古代文献的数据挖掘,发现古代中医认为本病的病位多见于肝,其次是肾,并且常常是肝肾同病;2、中医补肾方药对提前干预盐敏感性高血压有一定作用;3、盐敏感性高血压可以造成肾脏损害,且肾脏损伤主要表现为肾小管的损伤;4、高盐饮食造成了Ksp的表达异常。
