The model of animals with olfaction dysfunction has important application value for researches into olfaction-related diseases. Till now, there is no appropriate, objective and comprehensive olfaction assessment method for the model of animals with olfaction dysfunction. In this project, we choose rats that are infected by respiratory virus as the annimal model. And we take olfactory evoked potentials, olfaction system MRI and functional MRI as the examination methods for objective olfaction function test, take olfaction system protein and nucleic acid expression as the research approach to histology. Explore the establishment of the model of olfaction dysfunction steming from virus infection, related parameters of olfactory evoked potentials, olfaction system MRI and functional MRI, expression of olfactory marker protein(OMP)、matrix metalloproteinases(MMP) and tissue inhibitors of metalloproteinases(TIMP) in olfactory epithelium, expression of OMP 、MMP in olfactory bulb, c-fos protein expression in olfaction centrum as well as their relevance to function check. The projected results of this project are of important scientific significance to promoting basic research on olfaction-related diseases and realizing objective and comprehensive assessment of olfaction function. This will provide the basic method for examination of olfaction dysfunction positon and assessment of treatment effect. It is highly valuable for further research on olfaction dysfunction.
嗅觉障碍动物模型对于嗅觉相关疾病的研究具有重要的应用价值,但是到目前为止还没有合适的客观、全面的嗅觉障碍动物模型的嗅觉功能评价方法。本项目选用呼吸道病毒感染的大鼠作为动物模型,以嗅觉诱发电位、嗅觉系统结构磁共振成像和功能性磁共振成像为客观嗅觉功能检查方法,以嗅觉系统嗅标记蛋白、基质金属蛋白酶及其抑制产物、多巴胺和嗅皮层c-Fos蛋白表达作为组织学研究方法。探索病毒感染嗅觉障碍模型的建立,模型嗅觉诱发电位、嗅觉系统结构磁共振成像和功能性磁共振成像检查的相关参数;模型嗅上皮嗅标记蛋白、基质金属蛋白酶及其抑制产物表达,嗅球基质金属蛋白酶及其抑制产物表达和c-Fos蛋白在嗅觉中枢的表达,更为重要的是分析这些组织学变化和功能检查之间相关性。本项目预期结果对于促进嗅觉相关疾病的基础研究进展,实现客观全面的嗅觉功能评价具有重要的科学意义。
