In our study, animal model of polycystic ovarian syndrome(PCOS)in rats will be induced by the testosterone propionate(TP) and high-fat diet(HFD),insulin resistance ovarian granular cell model will be induced by dexamethasone. Based on collateral disease theory, rats will be intervened with acupuncture. The miR-21 expression profile will be examined by real-time RT-PCR and the expression of the key molecule protein PTEN, PI3K, Akt in PTEN- PI3K/Akt pathway will be checked by Western blot. Aimed at the miRNA-21 regulating PTEN/PI3K/Akt pathway as the breakthrough point, we will explore the molecular mechanism and target of acupuncture to improve PCOS ovarian insulin resistance. so as to provide scientific evidence for acupuncture therapy of insulin resistance in PCOS woman and to interpret the scientific connotation of acupuncture “to regulate collaterals of the uterus and menstruation and to pregnancy”.
采用丙酸睾丸酮及高脂饲料诱导PCOSIR的大鼠动物模型,采用低剂量地塞米松诱导大鼠颗粒细胞胰岛素抵抗的细胞模型,以络病理论指导针刺治疗作为干预手段,应用实时荧光定量PCR方法检测miR-21在PCOS颗粒细胞及卵巢中的表达,应用western blot方法检测PTEN/P13K/Akt信号途径关键分子PTEN、P13K和Akt蛋白的表达。旨在以miRNA-21介导PTEN/PI3K/Akt信号途径为切入点,探索针刺改善PCOS卵巢局部胰岛素抵抗的分子机制及作用靶点,将为针刺治疗PCOS合并IR提供实验依据,并有利于阐释针刺“调畅胞络,行经助孕”的科学内涵。
本项目按计划探索了针刺改善PCOS卵巢局部胰岛素抵抗的作用机制。本课题成功建立PCOS合并IR大鼠动物模型;筛选出PCOS合并IR大鼠卵巢组织差异表达的miRNAs表达谱,GO和KEGG分析预测其生物学功能;研究了针刺干预PCOS合并IR大鼠生殖和代谢变化,明确针刺能够改善PCOS代谢异常,筛选出针刺干预后PCOS合并IR大鼠卵巢组织miRNAs差异表达谱,揭示针刺改善PCOS卵巢局部胰岛素抵抗的可能机制,为针刺治疗PCOS合并IR提供实验依据,并有利于阐释针刺“调畅胞络,行经助孕”的科学内涵。
