In pathogenesis of prostatic diseases, epithelial barrier in acinus are often associated with destructure. However, some questions about balance of promoting permeability/keeping stability and difference in physiological/pathology situations are not clear. Combining with our previous studies, we posit that Musk-Olibanum have regulation patterns of “mutual reinforcement” to functions of prostatic epithelial barrier viewed from the “mutual” point and accorded with common characters of aromatic properties. We carry out the following researches around three regulation characters of “mutual reinforcement” including difference in physiological/pathology situations, balance of promoting permeability/keeping stability and barrier opening mechanism with selective specificity and cooperativity, in terms of key protein Claudins family of tight junction: 1) The hypothesis validation of Musk-Olibanum have regulation patterns of “mutual reinforcement” to functions of prostatic epithelial barrier; 2) The study on phosphorylation mechanism of Musk-Olibanum of regulation patterns of “mutual reinforcement” to functions of prostatic epithelial barrier; 3) The study on significance of Musk-Olibanum of regulation patterns of “mutual reinforcement” to functions of prostatic epithelial barrier in treatment of prostatic diseases. The study results will provide empirical evidence to support the “mutual reinforcement” compatibility of Musk-Olibanum and lay a substantial foundation for prostatic diseases treatment.
前列腺疾病均伴有腺泡上皮屏障功能损害。芳香药麝香-乳香对前列腺上皮屏障有促通透作用,但促通透性/维稳定性平衡,生理/病理状态下通透性差异等问题仍未探明。结合前期研究,根据麝香、乳香药性香窜的共同特点,我们从“相须”药效不可离的角度提出“麝香-乳香对前列腺上皮屏障功能‘相须’调控模式”假说。围绕上皮屏障功能“相须”调控三特点:生理/病理状态下的差异性调节机制,促通透性/维稳定性之间的平衡调整机制,以及选择特异性、协同性屏障开放机制,以紧密连接(TJ)蛋白Claudins为切入点开展以下研究:1)麝香-乳香对前列腺上皮屏障功能“相须”调控模式假说的验证;2)麝香-乳香对上皮屏障功能“相须”调控模式的磷酸化分子机制研究;3)麝香-乳香的上皮屏障功能“相须”调控模式在前列腺疾病中的作用与意义。本项目假说的验证,为理解麝香-乳香“相须”性效规律提供理论基础,及在前列腺疾病中的临床应用提供实验依据。
芳香药麝香-乳香“相须”为用,是促进前列腺上皮屏障通透性的重要原因。本项目根据麝香、乳香药性香窜,两者互相促进而“不可离”的共同特点,以“麝香-乳香对前列腺上皮屏障功能‘相须’调控模式”为切入点,围绕生理/病理状态下的差异性调节机制,促通透性/维稳定性之间的平衡调整机制,以及选择特异性、协同性屏障开放机制开展以下研究:1)麝香-乳香对前列腺上皮屏障功能“相须”调控模式假说的验证;2)麝香-乳香对上皮屏障功能“相须”调控模式的磷酸化分子机制研究;3)麝香-乳香的上皮屏障功能“相须”调控模式在前列腺疾病中的作用与意义。本项目通过深入研究麝香-乳香调节前列腺上皮屏障的精确分子机制,考察麝香-乳香介导 Claudind靶蛋白的上皮屏障功能调控模式在前列腺疾病治疗中的意义。从全新角度阐释和丰富了“相须”七情配伍理论的性效内涵,并为“芳香透窍”之麝香-乳香在前列腺疾病中的临床应用提供实验依据。
