Malignant gliomas are the most frequent malignant brain tumor in adults. Circular RNAs (circRNAs) are RNA transcripts that are widespread in the eukaryotic genome and play important roles in tissue development, gene regulation, and carcinogenesis. The clarification of circRNA regulatory mechanisms in glioma may be helpful to develop novel and effective target-therapeutic strategies. Previously we have performed circRNA microarry detection and found that circLIMD1 was significantly downregulated in glioma tissues compared with normal brain tissues, and low-level expression of circLIMD1 was significantly associated with a poor prognostic phenotype of patients with glioma. In addition, circLIMD1 could downregulate miR-372 and abrogate the oncogenic roles of miR-372, which may account for an important tumor suppressive mechanism of circLIMD1. On this basis, we will further investigate the role of circLIMD1, the possible mechanism of miR-372 regulation by circLIMD1 and the downstream target gene PHLPP2 of miR-372 in glioma progression. Meanwhile, we will determine the expression levels of circLIMD1 in a large number of glioma tissues, and analyze the relationship with clinical pathological characteristics and prognosis of glioma patients. Taken together, this study will provide a new perspective on circRNA and may contribute to novel and useful circRNA diagnostic and therapeutic targets for glioma.
胶质瘤是成人最常发生的恶性脑瘤。环状RNA(circular RNA, circRNA)大量存在于真核生物基因组中,并在组织发育、基因调控以及肿瘤发生中发挥重要作用。研究circRNA在胶质瘤中的重要调控作用有助于开发新型靶向性诊疗方案。我们前期对胶质瘤circRNA芯片筛查发现,表达异常下调的circLIMD1与胶质瘤病人不良预后密切相关。此外,circLIMD1能减少miR-372的表达并阻遏后者的原癌特性。本研究将在此基础上,深入研究circLIMD1在胶质瘤进展中的作用及circLIMD1对下游miR-372及其靶基因PHLPP2的调控机制。同时,我们将检测大样本组织中circLIMD1的表达,分析其与患者临床病理学特征及预后的关系。总之,本研究有望拓展我们对circRNA的认识,为寻找胶质瘤新的诊断及治疗靶标提供理论依据。
项目背景:胶质瘤是最常见的颅内恶性肿瘤。由于其高度浸润性,大多数患者无法治愈,总体生存率和治疗成功率都很低。因此,阐明胶质瘤的发病机制并寻找新的治疗策略至关重要。人类基因组绝大多数无蛋白编码能力,它们被转录成了非编码RNA(noncoding RNAs, ncRNAs)。已知ncRNAs不是RNA转录中的垃圾或噪音,其对基因表达有极其重要的调控作用,参与着发育、分化、增殖、免疫等几乎所有的生理和病理过程。近期研究表明,长链非编码RNA(Long noncoding RNAs, lncRNAs)在肿瘤中表达异常,影响着肿瘤的发生和发展进程。在胶质瘤中,已发现大量的lncRNAs发挥着重要的促癌或抑癌作用。LINC00346是位于染色体13q34上的一个基因间lncRNA,它的失调与多种癌症的进展有关。.研究内容:本研究旨在探讨LINC00346在胶质瘤中的功能作用和调控机制。1)分析163例胶质瘤组织样本和207例正常脑组织样本中LINC00346表达差异性,揭示LINC00346与患者预后的关系。对我院收集的20例胶质瘤组织和20例正常脑组织进行荧光定量PCR检测(qRT-PCR),分析LINC00346的表达及与病理分级的相关性。利用qRT-PCR技术检测LINC00346在正常人星形胶质细胞(NHAs)和神经胶质瘤细胞系(LN229、U251、H4和U87)中的表达。2)利用RNAi技术对LINC00346表达沉默,体外观察LINC00346下调对胶质瘤细胞增殖、凋亡、侵袭及迁移行为的影响。3)利用生物信息学软件预测、荧光素酶试验验证LINC00346与miR-340-5p、miR-340-5p与ROCK1的靶向结合关系。通过营救实验确定LINC00346调控miR-340-5p/ROCK1轴影响胶质瘤的进展。.重要结果:1)LINC00346在胶质瘤组织和细胞中呈高表达状态,且LINC00346高表达与胶质瘤患者的预后不佳及病理分级密切相关。2)LINC00346沉默对胶质瘤细胞生长和转移能力有抑制作用。3)LINC00346与miR-340-5p、、miR-340-5p与ROCK1均可以靶向结合。LINC00346通过与miR-340-5p竞争性结合进而调控靶基因ROCK1的表达,影响胶质瘤细胞的生物学进程。.科学意义:该轴可能有助于识别胶质瘤的潜在治疗靶点
