Genetic alteration is important hint for precise diagnosis and prognosis of biological behavior of malignant lymphoma. Bcl-6 is cloned from lymphoma and may have distinct expression pattern and be of important genetic alteration in lymphoma. The current subject aimed to study the expression pattern of reactive hyperplasia, different types of B cell and T cell lymphoma using immunohistochemistry. By using primers encompassing frequent region of 5' nonconding region of bcl-6 gene the microdissected tissue of different types of lymphoma were investigated for mutation, combined with FISH for rearrangement frequency. Bcl-6 expressed in nuclear in germinal center of lymphonode, majority of follicular lymphoma and DLBCL. Compared with expression of bcl-2 and CD10, bcl-6 could be another important marker of centroblast derived lymphomas. The mutation frequency of Chinese DLBCL was 18.4%, lower than that in the population of other countries. Bcl-6 might partly play a role in the lymphogenesis. FISH analysis is going on and the conclusion will be confirmed on more cases. The study will provide powerful evidence on molecular level for lymphoma classification and the mechanism of lymphoma development.
