The identification and functional characterization of the plant disease-resistance genes is one of the most important themes in creating excellent cultivar resistant to Colletotrichum spp., which will promote our understanding of the defense or resistance mechanism of plant. Anthracnose disease is a one of the most devastating diseases of persimmon (Diospyrus kaki L.) in orchards worldwide, and seriously hinders the development of the persimmon industry. Because of the high genetic diversity in Colletotrichum species, the lack of genome information and disease-resistant breeding in persimmon, the research of the molecular regulation mechanism and key genes during persimmon response to C.horii infection become much more difficult. In this project, Based on the analysis of genetic diversity and population structure of these Colletotrichum complexes isolates infecting persimmon. The expression profile of resistance (GS-6) stem infected with the major pathogen isolates of C.horii or not would be analyzed using the RNA-Seq technologies, respectively. Key resistance-related genes were obtained from several candidate genes through expression pattern analysis. Then the key resistance-related genes would be cloned and their function in plant-disease interaction through transient and stable transformation would be analyzed. The expected results will further our understanding on the molecular mechanism of persimmon resistance and provide new gene resources for persimmon breeding resistant to fungal diseases.
植物抗病相关基因的鉴定和功能分析是认识和了解植物抗感病机制的重要方面,且对优良种系的选育具有重要意义。柿树炭疽病是柿树上的毁灭性灾害,炭疽菌在生物学和遗传学上变异较大,柿树基因组信息少且抗病种质资源匮乏,致使柿树抗炭疽病相关基因的挖掘陷入瓶颈。本项目在分析了柿树炭疽病抗病品种“GS-6”的新梢响应柿树炭疽菌优势菌株(C.horii)侵染的表达谱,并筛选了多个候选抗病相关基因的基础上,通过表达模式分析进一步挖掘不同类型的关键抗病相关基因,利用亚细胞定位、瞬时表达和遗传转化等方面对抗病相关基因进行功能分析,初步探明不同类型抗病相关基因在柿树抗炭疽病过程中的作用机制。本项目在理论上可以深化对柿树抗病机理的了解,在实践上可为抗病分子育种提供优异基因资源,为病害防控提供新思路。
植物抗病相关基因的鉴定和功能分析是认识和了解植物抗感病机制的重要方面,且对优良种系的选育具有重要意义。柿树炭疽病是柿树上的毁灭性灾害。由于炭疽菌在生物学和遗传学上变异较大,柿树基因组信息少且抗病种质资源匮乏,致使柿树抗炭疽病相关基因的挖掘陷入瓶颈。本项目对我国柿树炭疽病病原种群进行了鉴定及致病力分析,获得了我国柿树炭疽菌的优势菌株,通过高通量测序获得了柿树炭疽菌优势菌株C. horii SD010基因组的完成图。并在此基础上利用转录组学技术分析柿树炭疽病抗病品种‘GS-6’的新梢响应柿树炭疽菌优势菌株(C.horii)侵染的基因表达谱,筛选得到21个候选抗病相关基因。对抗病相关NAC转录因子的表达模式进行了分析,发现NAC参与了柿果实的水杨酸和茉莉酸甲酯介导的抗病信号转导过程。深化了对柿树抗病机理的了解,为病害防控及抗病分子育种提供新思路。
