Bone marrow mesenchymal stem cells are an effective stem cell source for repairing tissue damage after pulmonary fibrosis, but the curative effect is limited, one of the reasons is that BMSCs, which live in the nests and survive in the lung, are few in number.Stromal cell derived factor 1 (stromal - derived factor 1, SDF - 1) and its specific receptors of chemokine receptor 4 (CXC chemokine receptor 4, CXCR4) consisting of SDF - 1 / CXCR4 axis in BMSCs homing and fixed value to participate in the repair of damaged parts play an important role in the process.Our previous studies showed that Maimendong Decoction effevtively improve pulmonary fibrosis in rats and which can promote BMSCs mobilization, homing as well as directional differentiation to alveolar epithelial cells.The purpose of this study is to explore BMSCs of Maimendong Decoction combined with Maimendong Decoction pretreatment to determine whether its role was performed through the sdf-1 /CXCR4 axis.
骨髓间充质干细胞(Bone Marrow Mesenchymal Stem Cells,BMSCs)是肺纤维化(Pulmonary Fibrosis)后修复组织损伤部分的一种有效的干细胞来源,但疗效有限,原因之一是归巢并存活于肺纤维的BMSCs数量较少。基质细胞衍生因子-1(stromal-derived factor-1,SDF-1)与其特异性受体趋化因子受体4(CXC chemokine receptor 4,CXCR4)构成的SDF-1/CXCR4轴在BMSCs归巢、定植到损伤部位参与修复的过程中发挥重要作用。我们的前期研究表明,麦门冬汤可以有效改善大鼠肺纤维化,且该作用可通过促进BMSCs动员、归巢以及向肺泡上皮细胞定向分化而实现。本研究旨在进一步探讨麦门冬汤联合移植麦门冬汤预处理的BMSCs是否通过SDF-1/CXCR4轴改善肺纤维化组织的修复。
研究背景:骨髓间充质干细胞是肺纤维化后修复组织损伤部分的一种有效的干细胞来源,但疗效有限,原因之一是归巢并存活于肺纤维的BMSCs数量较少,研究表明SDF-1/CXCR4轴在BMSCs归巢、定植到损伤部位参与修复的过程中发挥重要作用。本研究旨在进一步探讨麦门冬汤联合移植麦门冬汤预处理的BMSCs是否通过SDF-1/CXCR4轴改善肺纤维化组织的修复。重要结果及关键数据:麦门冬汤可增加肺纤维化损伤肺组织中SDF-1的分泌,使其高峰维持至纤维化后14天;麦门冬汤预处理可以提高BMSCs向纤维损伤肺组织的定向归巢和存活,促进肺组织炎症细胞浸润修复;麦门冬汤预处理的BMSCs的作用是通过SDF-1/CXCR4轴发挥的;不同浓度的麦门冬汤提取物对BMSCs均有增殖作用,以浓度为3.125mg/L干预24h细胞活性和增殖率最佳;麦门冬汤具有促进BMSCs迁移作用;麦门冬汤可上调BMSCsSDF-1 及CXCR4表达水平,提示麦门冬汤促进BMSCs迁移相关机制可能与麦门冬汤上调BMSCs SDF-1及CXCR4表达上调,激活SDF-1/CXCR4 生物轴相关。移植麦门冬汤预处理BMSCs能从减轻炎症、减少胶原沉积、抑制血管新生来改善肺纤维化;移植麦门冬汤预处理BMSCs治疗肺纤维化可能由novel-rno-miR-547-5p、rno-miR-3570、novel-rno-miR-406-5p调控,其中可能与肺纤维化密切相关的是Hippo信号通路、Notch信号通路、PI3K-Akt信号通路和Wnt信号通路。科学意义:本研究利用中药“多因微效”、多靶点、多效应、毒副作用小的特点,以前期基础研究的张仲景治疗肺痿经方麦门冬汤为研究对象,探讨了口服麦门冬汤联合麦门冬汤含药血清预处理能否进一步提高骨髓间充质干细胞的定向归巢和存活,进而探讨麦门冬汤联合移植麦门冬汤预处理的BMSCs是否通过SDF-1/CXCR4轴改善肺纤维化组织的修复,同时为寻找有效途径治疗肺纤维化打开新思路,并为同业人员临床治疗肺纤维化及用药提供实验基础。
