The roots of Dipsacus asper Wall. (Dipsacaceae) is a kind of staple Chinese herbal in traditional Chinese medicine, and a long history of clinical studies have shown their efficacy. Owing to very limited planting and heavy dependence on the wild D. asper resource, the reservation and biodiversity of wild D. asper were sharply reduced and the pesticide effect was variable. Germplasm is the major determinant for the yield and quality of medicinal herbs. Therefore, for the development of new cultivar with high yield and superior quality of Dipsaci radix, the collection, preservation, further evaluation and utilization of wild germplasm is extremely urgent. In this application project, our specific objectives are (i) to collect the germplasm widely and develop the species-specific SSR (Simple sequence repeat, SSR) marker used to genotype the germplasm, (ii) to evaluate the molecular genetic and phenotypic diversity of genuine in geo-authentic region for the collections based on population genetics, (iii) to determine the associations between whole genome polymorphism revealed by SSR and the phenotypic variations, including economic yield and its components, quality characteristics, (iv) to construct a core germplasm repository for D. asper, (v) to mining elite alleles and develop functional markers for molecular breeding, (vi) to elucidate the genetic and environmental factors underlying the genuine regional D. asper. Based on the study, we will provide a number of genetic resources for the genetic linkage map construction, key genes involved in the biosythesis of active ingredient and molecular breeding, and it further make an important contribution to the development for D. asper.
川续断根,又称续断,具有明显的药效和悠久的药用历史,为重要的大宗中药材。因长期掠夺式地采挖,导致川续断野生资源储备量及多样性锐减,因此其种质资源的收集、评价研究及新品种培育迫在眉睫。本项目拟在我国范围内广泛收集川续断种质资源的基础上,基于简化基因组测序开发物种特异、信息量丰富的SSR分子标记,对川续断种质资源进行基因分型并计算群体遗传学参数,考察道地产区恩施鹤峰种植条件下产量和品质等重要性状的表型变异,构建核心种质库;分析群体遗传结构,结合基因型数据对重要性状进行关联分析,探寻与重要性状关联的标记或基因位点,挖掘优异基因等位位点,基于等位位点的变异开发功能标记,解析川续断种质资源产量和品质等重要性状道地性形成的遗传基础和生态环境因子。该研究可为进一步开展川续断活性物质代谢途径关键基因克隆、遗传改良或分子育种等研究提供材料和理论依据,为川续断遗传资源的可持续利用及续断产业的稳定发展奠定基础。
