Results of research and experiments demonstrate that Dihuang Yinzi has effective on neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease etc.According to the TCM theory of Zang-manifestation"kidney stores essence ,nourishes marrow and communicates to the brain,which is sea of marrow",the view of kidney deficiency is the basic pathogenesis of senile diseases,and the modern medical knowledge of neurodegenerative diseases, this project,based on the previous work, pointing at neural stem cells, would use Fluorescent tags immunohistochemical method,flow cytometry, Real-time fluorescent quantitative PCR, Western blot ,and so on ,combining with the methods of cerebrospinal fluid pharmacology, to observe the influences of neural stem cell proliferation and differentiation, Wnt ,Notch and MAPK signaling pathways ,which are affected by classical ancient recipe Dihuang Yinzi for nourishing kidney-essence ,removing phlegm and kaiqiao. Study on mechanism of action of Dihuang Yinzi deep and systematically.Explore to use traditional Chinese medicine to impact and adjust the neurol functional reconstruction .Enriched modern scientific connotation of the TCM theory of Zang-manifestation"kidney stores essence and communicates to the brain".Provide important foundation for treating neurodegenerative diseases by nourishing kidney-essence ,removing phlegm and kaiqiao. Meanwhile,to lay the foundation for developing new drugs from Dihuang Yinzi ,which is famous for its effective on treating neurodegenerative diseases.
大量的研究报道和实验研究表明,地黄饮子对于神经退行性疾病如阿尔茨海默病、帕金森氏病等有较好的效果。本课题依据中医"肾藏精生髓,通于脑,脑为髓之海"的藏象理论,和老年疾病中肾虚为基本病机的观点,以及现代医学对神经退行性疾病的认识,在前期工作基础上,以神经干细胞为切入点,运用荧光标记免疫组化法、荧光实时定量PCR等技术手段,结合脑脊液药理学研究方法,观察具有补肾填精、化痰开窍功效的经典古方地黄饮子对神经干细胞增殖分化及对Notch信号通路的影响。对地黄饮子的作用机理展开深入系统的研究,探索应用中医药进行神经功能重建的干预调节机制,丰富"肾藏精生髓,通于脑,脑为髓之海"中医藏象理论的现代科学内涵,并为补肾填精、化痰开窍法治疗神经退行性疾病提供重要依据,同时也为治疗神经退行性疾病的有效名方地黄饮子的新药开发研究奠定基础。
本课题依据中医“肾藏精生髓,通于脑,脑为髓之海”的藏象理论,和老年疾病中肾虚为基本病机的观点,以及现代医学对神经退行性疾病的认识,在前期工作基础上,以神经干细胞为切入点,运用荧光标记免疫组化法、荧光实时定量PCR等技术手段,结合脑脊液药理学研究方法,观察具有补肾填精、化痰开窍功效的经典古方地黄饮子对神经干细胞增殖分化及对Notch信号通路的影响。研究结果显示,地黄饮子能促进神经干细胞的增殖分化,提高其抗氧化能力,且其作用机制与调节Notch信号通路有关。
