Parkinson's disease is a common neurodegenerative disorders characterized by the progressive degeneration of dopamine systems in substantia and striatum, and there is no effective therapy to address the cause of the disorder . Neural stem cell as a promising engineered cell provide an important source of cells for grafting into patients. We found the E14.5 rat mesencephalic progenitor cells can be expanded in EGF contained medium and retain its characteristics of self-renew and multipotency. The mesencephalic progenitor cells can be induced into TH immunoreactive neurons in the differentiated miedium containing conditioned medium of Mes42 or B49 cells, cell membrane fragments and cytokine IL-1, IL-11, LIF, and GDNF.IL-1 can immediately induce expression of Nurr1mRNA in mesencephalic progenitor cells and fail to induce Nurr1mRNA overexpression in striatal progenitor cells.Neural stem cell for transplantation in Parkinson's disease is in the progress.
建立神经前体细胞分离、培养、鉴定技术,观察神经前体细胞营养/生长因子受体表达及若干生物活性物质对其增殖、分化的影响,脑内移植后观察其迁移、分化规律。利用帕金森病大鼠模型探讨神经前体细胞作为基因修饰靶细胞在中枢神经系统疾病基因治疗中的应用价值。本课题旨在为神经前体细胞移植治疗中枢神经系统疾病提供理论依据和技术支持。
