针刺治疗AD的疗效已得到临床实践的证明,在相关基础研究中,我们发现内源性NSCs再生的激活、迁移和分化对于AD脑组织病损部位的修复、结构和功能重建,具有重要意义。前期的基础研究表明针刺能较好激发和调节内源性NSCs增殖和分化,但关于针刺促内源性NSCs迁移机制的动态研究及这一过程中NSCs分化发育的动态变化未见定论。本研究拟通过研究SAMP8小鼠经“三焦针法”干预后,可改善快速老化小鼠行为学,并且在不同干预组,不同时点SANP8小鼠脑组织中SVZa、RMS、OB 3个区域, SDF-1随针刺干预作用下增殖的神经干细胞增值、分化、迁移的时空表达情况,探究SDF-1的时空表达模式与针刺干预内源性增殖NSCs迁移对SAMP8大脑结构重建作用的相关性,完善“SDF-1在针刺治疗AD的促内源性NSCs增殖、迁移过程中具有积极影响”的假说,为针刺治疗AD进一步提供重要的理论和实验依据。
本实验基于SDF-1在NSCs迁移流中的时空表达,通过反映“迁移流”途径和方向的矢状面动态研究,探讨针刺对内源性 NSCs迁移这一神经再生修复过程必经关键环节的影响以及其动态变化,说明针刺促进内源性 NSCs 迁移和和分化作用对于神经再生修复、脑内结构和功能重建的重要意义。通过研究SAMP8小鼠经“三焦针法”干预后,观察快速老化小鼠行为学改善,检测针刺干预内源性 NSCs 增殖、并向成熟神经元、胶质细胞分化、发育的情况,以及随时间推移的动态变化,并且在不同干预组,不同时点SANP8小鼠脑组织中SVZa、RMS、OB 3个区域, SDF-1随针刺干预作用下增殖的神经干细胞增值、分化、迁移的时空表达情况,探究了SDF-1的时空表达模式与针刺干预内源性增殖NSCs迁移对SAMP8大脑结构重建作用的相关性。研究表明针刺能较好激发和调节海马和皮层内源性NSCs增殖和分化,分化形成成熟神经元,改善NSCs迁移流中的微环境。完善“SDF-1在针刺治疗AD的促内源性NSCs增殖、迁移过程中具有积极影响”的假说,为针刺治疗AD进一步提供重要的理论和实验依据。
