The study found that sphingomyelin from diet (mainly exists in egg, milk, and white meat) can inhibit colon cancer by regulating the key enzyme--alkaline sphingomyelinase (also called NPP7) ,which can hydrolyze sphingomyelin in intestinal mucosa. The effect that sphingomyelin inhibits colon cancer may be related to the balance of ceramide (tumor suppressor factor) and sphingosine-1-phosphate (S1P, carcinogenic factor) .But the mechanism is not clear. Our previous study showed that enhanced colonic tumorigenesis in NPP7 Knockout Mice with increasement of S1P in colon mucosa, suggesting that the tumorigenesis effect of S1P enhanced in NPP7 knockout mice. The aim of this project is:1) to find the change of sphingolipids and enzyme in NPP7 knockout mice;2)to explore that whether NPP7 inhibit colon cancer through Sphingosin kinase-1.
研究发现,可由饮食中提供的鞘磷脂(SM)通过上调其水解关键酶碱性鞘磷脂酶(NPP7)的表达而发挥其抑制结肠肿瘤的作用,该作用可能与SM在代谢过程中产生的神经酰胺Cer(抑癌因子)与磷酸鞘鞍醇S1P(促癌因子)之间的平衡和消长有关,但是NPP7如何调节二者之间的平衡仍不详。本课题组发现,NPP7基因敲除小鼠结肠粘膜中促癌分子S1P明显升高,其结肠肿瘤的成瘤率和瘤癌变率显著升高,提示NPP7参与结肠肿瘤的发生发展。本研究拟采用NPP7基因敲除小鼠制备结肠肿瘤模型,比较药物干预前后结肠粘膜中Cer/S1P的变化,以及与S1P生成相关的关键酶Sphk1的表达差异;从动物与细胞水平,明确NPP7可通过影响Sphk1的活性,调节Cer/S1P二者关系,从而发挥其调控结肠肿瘤产生的作用。该研究将为进一步探索治疗结肠癌的靶点药物奠定理论基础。
