Malignant tumor is a major threat to human health, the important biological characteristics of invasive growth and metastasis, and its formation and metastasis is a common action of multiple genes, multiple factors.Therefore, to explore and find the occurrence or metastatic mechanism of tumor, is one of the research topics of basic and clinical oncology.How to prevent malignant tumor recurrence and metastasis is a hot topic in the field of medical research.Our previous study found that, by using low-frequency, high mechanical index of ultrasound mediated microbubble destruction enhanced cavitation effect, can cause the number and tumor micrometastasis size decreased significantly, but the mechanism still needs further study.This study intends to adopt the different sound intensity ultrasonic, along with the irradiation time and increase the mechanical index, the use of ultrasound mediated microbubble destruction targeting, by experimental study and theoretical analysis of in vitro and in vivo:ultrasonic irradiation time, mechanical index in ultrasound mediated microbubble destruction enhanced cavitation effect are the most important factors, the hypothesis and have important effects on the cytoskeleton. To find out the optimum irradiation conditions, aggregation state of F-actin expression in the liver metastasis of colon cancer cell skeleton and antisense cortactin detection, can be expected to improve curative effect of comprehensive cancer treatment provides a new means and experimental basis.
恶性肿瘤是威胁人类健康的重大疾病,其重要生物学特性是侵袭性生长和转移,而其形成和转移是一个多基因、多因素共同作用的结果。因此,探讨和寻找肿瘤发生或转移的机制,是肿瘤学临床及基础研究的主题之一。如何预防恶性肿瘤的复发和转移是医学研究领域的热点。我们前期研究发现,利用低频、高机械指数超声介导破坏微泡增强空化效应,能够引起肿瘤微小转移灶的数目及肿瘤的大小明显减少,但对其机制尚且有待于进一步研究。本课题拟采用不同声强超声波,随着超声辐照时间以及机械指数的增加,利用超声介导破坏微泡的靶向作用,通过体外及体内的实验研究和分析:超声波的辐照时间、机械指数在超声介导破坏微泡增强空化效应中均是其重要影响因素,并对其细胞骨架有重要的影响"的这一假说,摸索最佳的辐照条件,检测结肠癌肝转移中细胞骨架的聚合态F-actin及反义皮层肌动蛋白的表达,可望为提高肿瘤综合治疗的远期疗效提供新是手段及实验依据。
