Homology of medicine and food has a long application in Chinese ancient culture, pharmacodynamic research of traditional Chinese medicine is closely related with gastric and gastrointestinal microbiota.Studying the correlationship of traditional Chinese medicine and probiotics on host microecology and infection at macro- and micro-levels has become a basic research focus of modernization of traditional Chinese medicinal and edible materials. Up-to-date, we still show little evidences and knowledge on the Helicobacter pylori-induced gastric microecology, moreover, using 16S rDNA high-throughput sequencing approach to investigate the microecological transition and relative intervention strategies by HP infection in stomach was still not clearly analysed and documented. Hence, basing on our former experiment that probiotics and medicinal & edible mateirals can jointly improve HP-induced gastric inflammation in mice, in this study, we will adopt lactic acid bacteria and Allium chinense G.Don for testing the respective and joint effects on HP infected mice model. Further investigation i.e. 16S rDNA high-throughput sequencing method, immunohistochemistrical staining, microbial analysis etc. will be performed to get a well understanding of HP-induced gastric microecological transition and the correlation mechanisms of united exposures to probiotics and Chinese medicine for prevention and treatment of HP infection, and finally provide a scientific research opinion or basis for prevention and treatment of HP-induced gastric inflammation.
药食同源,传统药食两用中药(食材)的功效研究与胃肠道微生态菌群的变迁和代谢活动密切相关。从宏观、微观两个角度,考察药食两用食材与益生菌对宿主微生态群系的变迁和机体感染炎症改善的关联性,已成为药食同源产业现代化的基础研究热点问题之一。迄今,人类对幽门螺杆菌感染引起的胃部微生态环境的认识仍知匮乏,尚未见利用高通量16S rDNA测序技术,探究其微生态菌群变迁与改善胃炎炎症相关性的研究。本项目在前期获得的益生菌和江中健胃消食片改善幽门螺杆菌感染小鼠炎症的基础上,采用乳酸菌、药食两用食材-藠头为原料,利用高通量16S rDNA测序、免疫组化、微生物群系分析等技术手段,通过幽门螺杆菌感染的小鼠模型,阐述幽门螺杆菌感染引起的胃部微生态群系变迁,以及用益生菌和藠头预防和改善幽门螺杆菌感染的(协同)作用机制,为指导防治幽门螺杆菌引发的胃炎提供新的思路,为丰富中药微生态学理论提供实验依据。
药食两用食材、益生菌的健康作用与胃肠道微生态菌群的变迁和代谢活动密切相关。迄今,由幽门螺杆菌感染引起的胃部微生态环境的变迁方面的认识仍很匮乏,尚未见利用高通量16S rDNA测序技术,探究其微生态菌群变迁与改善胃炎炎症相关性的研究。.本文从体内和体外角度,以及细胞水平,考察了酸豆角来源的植物乳杆菌ZDY2013对小鼠胃肠道微生态的影响规律,对其预防幽门螺杆菌感染的实际效果进行了探究。重要结果有:植物乳杆菌ZDY2013可以体外有效抑制幽门螺杆菌的生长,且有较强的黏附胃腺癌细胞AGS的能力,能抑制幽门螺杆菌对AGS的粘附作用;植物乳杆菌ZDY2013可以有效抑制由幽门螺杆菌诱导的TNF-α转录水平的表达,提高抗炎症因子IL-10的转录表达水平。HE染色结果表明,植物乳杆菌ZDY2013能预防幽门螺杆菌感染小鼠引起的胃粘膜炎症反应;16S rRNA基因高通量测序结果表明,ZDY 2013灌胃能有效地预防幽门螺杆菌感染及其引起的胃微生态改变。此外,植物乳杆菌发酵可以提高藠头的生物功能,营养价值和化学性质;植物乳杆菌ZDY2013极端耐酸,其极强的黏附能力和拮抗致病菌的能力与菌体的表层蛋白密切相关。.上述研究结果将为利用益生菌和药食两用食材-藠头的益生作用,防治幽门螺杆菌引发的胃肠道微生态变迁及抗炎症,提供了新思路,为丰富中药微生态学理论提供了实验依据。
