In the light of the morbidity and mortality of lung cancer are among the top of the malignant tumor, the further research about the development and metastasis and individual therapy with low toxicity of lung cancer is an important topic which is so urgent to solve in lung cancer research. The EMT process is recognized as one of the mechanisms of tumor metastasis. But the mechanism how lncRNA regulate EMT is not clear. Our team used TGF - beta to induct the non-small cell lung cancer cell line A549, and successfully built a EMT model. On the basis of using chip screening, we found that with the occurrence of EMT, a lot of lncRNAs expression showed a trend of rising or falling. The phenomenon prompts the lncRNA with obvious changes may play an important role in the process of EMT, and may provide research foundation for further exploring biomarkers about EMT and targets of treatment. This topic proposed one of differential LncRNA----LncRNA-TBDR12 as the research object. We verified the consistency of TBDR12 with the results of the chip.This project want to reveal the role of lncRNA-TBDR12 in the lung cancer development through lots of assays, such as transfection assay、gene knock-down assay、Pull down assay、RIP assay. We hope that we could find a useful biological marker related with the migration and invasion of lung cancer cells and provide a theoretical basis for the clinical treatment.
肺癌具有高侵袭转移性,这一特点使其死亡率居恶性肿瘤之首。EMT过程是公认的肿瘤侵袭转移的机制之一。LncRNA由于参与了生物体内多种生理病理过程,已成为研究的热点,但其调控EMT过程的机理尚未明确,还有待于进一步深入。本课题前期利用TGF-β诱导非小细胞肺癌A549细胞系,构建EMT模型,在此基础上利用芯片检测发现了一些具有调控EMT潜能的LncRNA。经过筛选及验证,选定LncRNA-TBDR12为研究对象。前期研究发现LncRNA-TBDR12在EMT过程中变化显著,这提示LncRNA-TBDR12可能通过调控EMT在肺癌的侵袭转移过程中发挥重要作用。本课题利用转染、基因敲降、q-PCR、RNA pull-down、RIP等方法对LncRNA-TBDR12在肺癌EMT过程中的潜在作用及机制进行研究,以期为肺癌侵袭转移机制的研究提供理论依据,为临床肺癌侵袭转移的预警和治疗提供新的靶点。
