Podophyllotoxin;Hypoxanthine phosphoribosyltransferase;GE mutation.By means of cell clone methods HPRT gene mutation frequence in 97 health children volunteers of shanghai city was investigated who were divided into three groups depending on their age, it was found that there is correlative between enviroment factors and progress of population "Carcinogensis" probability. The technology process to detect HPRT gene mutation was estabilished ,it has offered some importment condition to set up basic value of HPRT gene mutation frequence inchinese children in a large population. Detecting those children's HPRT gene mutation frequency before and after chemotherapy, it was confirmed that the method is effective in investigating impairment degree of patient's gene and predicting occurrence of secondary tumor. It means that detecting risk degree of human cell gene damage in leukemia children after chemotherapy with different protocolm but same effect, It is helpful for modifying component in chemotherapy program to research the mechanism of secondary tumor from epipodophyllotoxin.
通过多种分子生物学技术,分析各年龄阶段中国儿童HPRT基因突变频率和分布状况,可望发现新的基因突变类型;结合药代动力学研究,观察表鬼臼毒素累积对该基因突变频率的动态影响,寻找该类药物导致的体细胞基因严重损伤的危险因子,为探索肿瘤发生机制、临床合理使用鬼臼毒素,以及避免继发性急性白血病和骨髓异常增生综合症发生提供有效证据。
