Extracts of Senna and Croton, which were in certain range of dosage, can enhance clearly gastrointestinal motility by different pathways of administration with cathartic effect, gastrointestinal propellant movement experiment, X-ray analysis and morphological changes of gastrointestinal tissues. Long term of gastric administration of the extracts of Senna and Croton induce inflammatory changes and cell damage in the whole layer of whole gastrointestinal tract. Effect of extracts of them on intestinal epithelia cells and smooth muscle cells were investigated by MTT,flow cytometer, immunohistochemistry,Dot blot and DD-PCR. The results show that intestinal epithelia cells growth and proliferation were inhibited in a dosage-dependent manner by extracts of them. The malignant conversion of the cellular phenotype was observed. They have direct effects on the contractility of Guinea pig colonic smooth muscle cells. 18 differential proteins induced by Croton were isolated from intestinal tissue of model mice and 21 differential proteins induced by senna were isolated from colon of model mice using 2-D PAGE, and the amino acid sequence at N-terminus was determined in some of them. The differential proteins screened from the intestinal tissue of model mice might mediate the enhancement effects of senna and croton on gastrointestinal motility.
本研究拟用导泻中药巴豆和番泻叶粗提物灌胃制备胃肠运动亢进动物模型,双向SDS-PAGE法分离并比较正常与模型动物胃肠组织中蛋白质电泳图谱的差异,从胃肠道组织中寻找并分离出这两种中药诱导的、与胃肠运动亢进相关的蛋白质,进一步用RLM-RACE法克隆出其基因,为用这些分子及基因设计和研制高效低毒的胃肠动力新药奠定基础。
