p,p'-DDE is one of the persistent organic pollutants, which has significant adverse effects on human reproductive function. So far, there is no effective way to antagonize its reproductive toxicity. Previous studies have found that the dysfunction of oocyte oxidation / reduction regulation results in the development potential of oocytes in women with Phlegm Dampness syndrome infertility, which is similar to the reproductive toxicity mechanism of p,p'-DDE.Chinese medicine has good effects on Phlegm Dampness syndrome infertility, and the mechanism may be regulating the abnormal function of oxidation/reduction regulation.Therefore, we raise the hypothesis that “p,p'-DDE is likely to amplify the disturbance of oxidative stress in Phlegm Dampness syndrome infertility, Qi Gong Wan may regulating oocyte antioxidant pathways and antagonizing its reproductive toxicity".The present study will use Phlegm Dampness syndrome model rats and oocytes to investigate the the antioxidant effect and the antagonism of traditional Chinese medicine of Qi Gong Wan, in order to provide evidence for the use of Chinese herbal compound to reduce the adverse effects of environmental pollutants on the human.
p,p'-DDE是持久性有机污染物,对人类生殖机能有显著不良影响,目前尚无有效拮抗其生殖毒性的方法。前期研究发现,卵细胞氧化/还原调节机能异常造成痰湿型不孕症女性卵子发育潜能受损,与p,p'-DDE生殖毒性机制相似。中药复方对痰湿型不孕症疗效明确,生物学基础可能是调节氧化/还原调节机能异常。因而提出“p,p'-DDE可能对痰湿证患者体内的氧化应激起到放大的作用,治疗痰湿型不孕症经典方剂-启宫丸,可能通过调节卵细胞内抗氧化通路进而对p,p'-DDE生殖毒性起到拮抗作用”假说。本研究拟采用痰湿证模型大鼠及卵细胞体外培养的方法,进行启宫丸对痰湿证模型大鼠卵子细胞氧化应激、发育潜力及生殖毒性拮抗作用的研究,从中药抗氧化作用及纠正p,p'-DDE不良影响两方面阐明启宫丸治疗痰湿型不孕症及拮抗p,p'-DDE生殖毒性的作用和机制。为使用中药复方降低环境污染物对机体的不良影响提供证据支持。
