Diabetes mellitus is one of the risk factors of periodontitis. The inflammation cytokine upregulation by high glucose level is considered to be a major reason for periodontal tissue destruction. Visfatin, an adpokin which is secreted by adipocyte, is correlated with plasma glucose level. It also can be detected in periodontal tissue. One of the function of visfatin is inflammation regulation. It can upregulate cytokine production expression by NF-κB signal pathway. So, we guess that: periodontal tissue destruction on diabetes is supposed to be mediated by visfatin. This study contain five parts: (1) visfatin expression in salive, gingival crevicular fluid and periodontal ligament on patients with diabetes and periodontitis; (2) the effect of different glucose level on visfatin expression in periodontal ligament cells; (3) the effect of visfatin on inflammation cytokines expression in periodontal ligament cells; (4) NF-κB signal pathway for visfatin regulation mechanism on inflammation cytokines expression; (5) visfatin expression in vivo and protection effect of visfatin inhibitor on periodontal destruction. This study plan to find the inflammation mechanism that diabetes affect periodontal destruction, and more to guide the treatment study.
糖尿病是牙周炎的危险因素之一,目前认为糖尿病导致牙周组织炎症因子释放增加,加重了牙周组织的破坏。visfatin是一种内脏脂肪素,其表达分泌与血糖呈正相关;牙周组织中能够检测到visfatin。visfatin 同时能够通过NF-κB上调炎症因子表达。因此,我们猜想:糖尿病加重牙周炎组织破坏可能是通过visfatin介导。研究内容:1.visfatin在伴糖尿病牙周炎患者机体的表达及其与血糖和牙周炎的相关性;2.不同糖浓度对牙周膜细胞visfatin表达的影响;3.visfatin对牙周膜成纤维细胞炎症因子表达的影响;4. NF-κB信号通路在visfatin影响PDLC炎性因子表达调控机制;5.体内验证visfatin表达分布及visfatin抑制剂对牙周组织炎症的拮抗作用。本项目拟通过上述方案研究糖尿病影响牙周炎的炎性机制,并为治疗提供新的靶点。
