Abutilon indicum (L.)Sweet, is a traditional minority medicine from Guangxi province. This herb have efficacies of displling wind and clearing heat, benefit qi for resuscitation, dissipating phlegm for diuresis, which can be used for treatments such as cold fever, mumps, gonorrhoea, Tinnitus and deafness, Hernia carbuncle swollen and so on. Preview research from this project group have show that antiinflammatory action was observed in Ethanol part extract in Abutilon indicum (L.)Sweet, diuresis function in petroleum ether extract as well. In this study, technologies and methods such as chemistry of Chinese materia medica, pharmacology of traditional Chinese medicine, serum pharmacochemistry and LC-MS Analysing are used to comfire the active ingredients of effective extract in the serum of experimental animals; combined with the test of swelling rate that cause by dimethylbenzene or carrageenin and swelling depressive rate to explore the conection between anti-inflammatory action of Abutilon indicum (L.)Sweet and activation of hypothalamus-pituitary- adrenal cortex axis. Datas of numeration of leukocyte, contents of protein, PGE2, NO, MDA, TNF-α, IL-1β, IL-6, activity of LBT4, SOD, MPO will be test in order to study the anti-inflammatory mechanism and contents of Na+, K+, Cl-, ADH, AngⅡ, ANP and ALD in rat's urine are test to investigate the Diuretic mechanism of Abutilon indicum (L.)Sweet.
磨盘草Abutilon indicum(L.)Sweet为广西民族药材。本品具有疏风清热、益气通窍、祛痰利尿等功效,用于感冒高烧不退,流行性腮腺炎、淋病、耳鸣耳聋、疝气痈肿等症。课题组前期研究表明:磨盘草乙醇提取物具有明显抗炎作用;石油醚提取物具有显著利尿作用。本课题拟运用中药化学、中药药理学和中药血清药物化学技术和方法,以LC-MS跟踪分析,确定磨盘草有效部位提取液在实验动物血清中的移行成分;结合二甲苯、角叉菜胶对所致实验动物肿胀与肿胀抑制率的测定,探索药材抗炎作用是否与活跃下丘脑-垂体-肾上腺皮质轴有关;同时检测实验动物白细胞计数、蛋白含量、 PGE2、NO、MDA、TNF-α、IL-1β、IL-6的含量及LBT4、SOD、MPO的活性等多项实验指标,确定其抗炎作用机制;拟通过检测大鼠尿液中Na+、K+、Cl-以及ADH、AngⅡ、ANP和ALD的含量,探索药材的利尿作用机理。
磨盘草Abutilon indicum(L.)Sweet为广西民族药材。本品具有疏风清热、益气通窍、祛痰利尿等功效,用于感冒高烧不退,流行性腮腺炎、淋病、耳鸣耳聋、疝气痈肿等症。本课题运用中药化学、中药药理学和中药血清药物化学技术和方法,确定了磨盘草石油醚部位和乙酸乙酯部位具有利尿作用,正丁醇部位具有明显抗炎作用;建立了抗炎利尿有效部位的指纹图谱。磨盘草石油醚部位可改善异丙肾上腺素所致的大鼠心衰,其机制可能与降低血清中的ANP、AngⅡ、Ald的水平相关。磨盘草乙酸乙酯部位能减少肝硬化腹水大鼠的腹腔积液和降低ALT和AST的水平,并能改善肝功能。磨盘草正丁醇部位对急、慢性炎症都有一定的抑制作用,其抗炎作用可能与通过抑制白细胞浸润和游走、减少渗出、消除氧自由基、减少炎症介质PGE2合成及抑制TNF-α、IL-1β生成,兴奋下丘脑一垂体一肾上腺皮质轴等有关;对佐剂性类风湿性关节炎模型大鼠具有一定的疗效,其作用机制可能与IL-1β、IL-6水平有关。本项目所建的指纹图谱方法简单,准确可靠,重复性好,可为后续谱效关系研究提供一定的依据。
