The Tan Sheep is the unique precious fur sheep breeds, the product of Ermao fur from Ningxia Tan Sheep has a high reputation in the domestic and international fur market. However,the molecular mechanism of the Ermao fur formation is still unclear. It is well known that the regulation of gene expression plays key role in the process of the growth and development of clothing hair. And miRNA regulates the developmental processes of skin follicles in mammalian in the post-transcriptional level. In the proposed research, we will focus on the skin and hair follicles of Tan Sheep and Han Sheep. In order to screen the Tan Sheep follicle-specific expression of miRNA, and further predict and identify the specificity target genes of miRNA at different developmental stages of skin and hair follicle of Tan Sheep, we will assess the characteristics of miRNA expression in the different developmental stages of Tan and Han Sheep skin and hair follicle by means of high-throughput sequencing combining the Bioinformatics analysis. Meanwhile, the localization and quantitative expression of the candidate genes which were regulated by the Tan sheep candidate miRNA are characterized in cellular and molecular level by using the methods of the tissue sections in situ hybridization, immunohistochemistry. This proposal will eventually clarify the molecule regulated mechanism of the key miRNA in the development process of skin and hair follicle, and reveal the impacts on the growth and development of skin and hair follicle in Tan Sheep, may provide a new theoretical basis for the breeding, fur quality and efficient production of Tan Sheep.
滩羊是我国特有的裘皮用绵羊品种,尤其是宁夏滩羊所产的"二毛裘皮"在国内外毛皮市场上享有较高的声誉。但目前关于滩羊"二毛裘皮"形成的分子机制还未阐明。已知基因表达调控是被毛生长发育的决定性因素,且miRNA在转录后水平上调控着哺乳动物皮肤毛囊的发育过程。基于此,本项目拟以不同发育阶段的滩羊与小尾寒羊皮肤毛囊为实验材料,利用高通量测序技术结合生物信息学方法,分析两个品种间不同发育时期皮肤毛囊中miRNA表达谱特征,筛选出滩羊皮肤毛囊特异性表达的miRNA,预测并鉴定其靶基因;同时利用原位杂交、免疫组织化学等方法从细胞和分子水平上系统的分析滩羊特异性miRNA调控的靶基因在组织中的定位与定量表达;进而阐明特异性miRNA在滩羊皮肤毛囊发育过程中的分子调控机制,揭示其对滩羊皮肤毛囊生长发育的影响。本项目的实施为快速、准确选育优质高产滩羊品系提供分子标记,也为人工调控裘皮的高效优质生产提供技术支撑。
基因调控是被毛生长的决定性因素。从基因水平上揭示绵羊被毛生长的特点,对于改良其品质具有重要的意义。miRNAs在调控基因表达方面起重要的作用。基于此,本项目通过高通量测序和生物信息学分析技术对刚出生、出生30 d和成年期3个不同发育阶段的滩羊和小尾寒羊皮肤毛囊组织中的miRNA表达进行分析。在此基础上,比较了同一品种不同发育时期皮肤毛囊中miRNA表达的差异,以及不同品种相同发育时期miRNA表达的差异。最终在滩羊和/或小尾寒羊皮肤毛囊中筛选出大量差异表达的miRNA。对差异表达miRNA进行靶基因的预测及验证,分析特异性miRNA及其调控的靶基因在绵羊皮肤毛囊发育中的调控作用。主要研究结果如下:.(1)在小尾寒羊3个不同发育时期皮肤毛囊组织中共鉴定出1062个miRNAs,包括160个已知miRNAs和902个新发现miRNAs;其中226个为差异表达miRNAs,靶基因预测共得到2645个靶基因。.(2)在成年期滩羊和小尾寒羊皮肤毛囊组织中共鉴定出561个miRNA,包括138个已知miRNA和423个新发现miRNA;其中79个为差异表达miRNA,并对其靶基因进行预测分析。.(3)生物学重复测序后在出生30 d和刚出生滩羊皮肤毛囊组织中共筛选到43个差异表达的miRNA。对其中的oar-miR-433-3p进行靶基因验证和分析,通过双荧光素酶报告基因系统证实FGF7为其调控的靶基因。最后通过实时荧光定量PCR和Western Blot方法分析了FGF7基因在不同发育阶段滩羊皮肤组织中的表达,结果证实相对于刚出生滩羊而言,出生30 d时滩羊皮肤毛囊组织中的FGF7表达显著升高(P<0.05)。据此推测oar-miR-433-3p可能通过调控其靶基因FGF7在不同发育阶段滩羊皮肤毛囊中的表达,最终参与了滩羊皮肤毛囊的发育,具体的调控机制有待于进一步的研究。. 本项目的实施为在miRNA水平上解析滩羊“二毛裘皮”优良性状形成的分子机制提供了理论依据,同时也为选育优质裘皮用绵羊品种提供了分子标记。项目的进一步研究可为最终通过转基因技术培育优质裘皮用绵羊品种提供技术支撑。
