Chronic wound is the clinical common disease, which has become an important factor of harm to people's health due to lack of effective therapy in clinic. It is the hotspot and difficulty of modern medical science research to find new therapeutic targets and strategies. Researches show that PTEN plays an important role in cell growth, proliferation, differentiation, apoptosis, adhesion, migration and angiogenesis, also it participates several signalling pathways in wound healing. TCM treatment in skin ulcer has a long history, the effect is obvious. MEBT/MEBO is originated from removing slough and promoting growth of tissue regeneration. Research group previous studys find that MEBT/MEBO could improve the ultrastructural of intracellular organelles, maintain a normal intracellular homeostasis, Accelerate bFGF、VEGF proliferation and differentiation, but the mechanism is not yet fully clear. On this basis, this research uses that method in treating chronic wound model in rats, adopts techniques of Western Blot, qRT-PCR and ultramicroscopic pathology to discuss the wound healing molecular mechanism of MEBT/MEBO through PTEN/PI3K/Akt and PTEN/Akt/β-catenin signaling pathways. It can provide effective treatment and theoretical foundation for treating chronic wounds by using MEBT/MEBO in clinical treatment.
体表慢性难愈合创面是临床常见病症,目前缺乏有效的治疗手段,已成为危害人类健康的重要因素。探求新的治疗靶点及策略已成为现代医学研究的热点与难点。研究表明PTEN在细胞生长、增殖、分化、凋亡、黏附、迁移和血管生长中起着重要作用,参与了创面愈合的多条通路。中医药治疗皮肤溃疡历史悠久,效果明显,皮肤再生医疗技术(MEBT/MEBO)源于中医祛腐生肌法。课题组前期研究发现MEBT/MEBO能明显改善大鼠表皮生长细胞超微结构,稳定细胞内环境,加速bFGF、VEGF的增殖分化,但机制尚未完全清楚。在此基础上,该研究通过对慢性难愈合创面模型大鼠应用该法治疗,并运用Western Blot、qRT-PCR、超微电镜等技术,从PTEN/PI3K/Akt和PTEN/Akt/β-catenin信号通路探讨该医疗技术促进慢性难愈合创面修复的分子机制,为临床应用该技术防治慢性难愈合创面提供有效的治疗方法和理论依据。
