Preliminarey attempts to purify the autoregultor for thiemamycin biosynthesis from Streptomyces cattleya were conducted. During the research of the physiological regulation of thiemamycin biosynthesis,nitrates were found to be able to promote the production of thienamycin, and ammoniums, on the contrary, decrease thienamycin production levels. A 900bp DNA fragment from the genomic DNA was cloned and sequenced. Sequence analysis indicated that it had a complete ORF encoding some Ser/Thr kinase,highly suggesting that this ORF product participate the regulation of antibiotic (thiemamycin) production.
研究硫霉素生物合成调控的机理,从卡特利链霉菌原株发酵液中分离纯化调节因子,测定调节因子的化学结构,同位素标记调节因子,从卡特利链霉菌原株细胞抽提物中分离纯化调节因子结合蛋白,测定其氨基酸序列,制备核苷酸探针,克隆编码调节因子结合蛋白的基因,进行基因阻断实验,获得因子结合蛋白缺陷变株,分析调节因子及其结合蛋白调控的机理。.
