Conjugated nitrene due to the interaction of pi orbital and p orbital of Nitrogen, could exhibit 1,3-dipole nature. Usually, these nitrene intermediates are too reactive and show poor selectivities in reactions. Transition metals have lots of excellent properties, and should render the conjugated metal nitrene reaction highly selective and under mild conditions. From proper nitrene precursor, under the help of organic metal, conjugated metal nitrene could be generated and intercepted in situ by alkene or alkyne to give five membered azo-rings such as pyrole/pyroline, imidazole/imidazoline, oxazole/oxazoline, thiazole/thiazoline and so on. These would address the problems encounter by conjugated nitrenes. These new [3+2] cycloaddition reactions will not only enrich the nitrene chemistry and our understanding of these reactive intermediates, but also provide new strategies for medicinal synthesis. So this program is highly valuable in terms of both academic and industry.
共轭氮宾由于轨道的相互作用可以展现为1,3-偶极体的反应性。这些活性中间体非常活泼,其反应选择性往往比较差。过渡金属的许多优良特性使得这一类共轭金属氮宾的反应具有高选择性,条件温和等特点。从相应的氮宾前体出发,在过渡金属的存在下现场生成金属氮宾/1,3-偶极体,并用合适的亲偶极体比如烯烃和炔烃对这些中间体进行现场捕获,可以高效的构筑五元含氮杂环比如吡咯/啉、咪唑/啉、噁唑/啉、噻唑/啉等。这一类新的【3+2】环加成方法学的发展将不但丰富了金属氮宾化学,加深对这一类反应中间体的理解,也会为药物合成提供新的合成策略。也即该项目的实施既有学术意义具有实用价值。
在本基金的资助下,我们课题组开展了对共轭氮宾1,3-偶极体反应性的探索。采用在适当亲偶极体存在下,现场捕捉新生成的氮宾的策略。我们探索了数种反应条件,特别是试剂/催化剂组合,虽然没有观察到共轭氮宾与烯、炔烃的[3+2]环化产物,但发现了一些新的化学反应,主要发展了三类非常有用的反应。(1)1-磺酰三氮唑取代的胺衍生物发生各种卡宾参与的插入、环化再重排等反应,获得各种氮杂环;(2)烯基炔胺与磺酰叠氮反应得磺酰烯亚胺中间体,分子内先环化再[3, 3]重排得环状脒类化合物;(3)以烯基叠氮为前体的氮杂环丙烯与分子内1,3二烯体的[4+2]环加成反应,获得氮杂5-6-3或6-6-3并环系统。这些探索性工作为今后的研究提供了依据,明确了将来的研究方向,也产生了几种高效制备结构多样氮杂环的新策略。该项目的实施既产生了一些领先的学术成果,也产生了具有相当不错实用价值的成果。
