An attenuated rabies vaccine (SRV9 )and LT (heat-labile toxin derived from E coli) were inoculated mice by direct oral cavity instillation, stomach delivery and rectal instillation, respectively. The immune response and the role of LT were evaluated based on detection of specific lymphocyte proliferation in vitro, cytotoxin T cells response, titer of serum IgG and IgA, mucosal secretory (SIgA), neutralization index of serum antibody and protection rates after challenge, The results showed that the immune response evoked by rectal instillation were higher than others were. The immune responses in groups with LT were higher than without LT; Some biogradable materials such as gelatin, walnut oil and polybutylcyanoacrylate(PBCA) were chosen to encapsid the attenuated rabies vaccine, microspheres of different sizes were prepared and inoculated the mice by oral or i.m. inoculation. The data demonstrated that the encapsided microspheres can't stimulate protective immune response by either ways; Rabies virus and nucleoprotein gene were detected by histochemical stain under the electron microscope and by RT-PCR 6 hrs after oral delivery , neither antigen nor gene was detected in digestive tract; some material were tested for their delay characteristic in attenuated rabies vaccine, a stabilizer formula (including peptides, newborn cattle serum ,glycerol and EDTA )was found out, and a standard ELISA kit for detection the antibodies of dogs was also exploited.
