alpha-amylase gene from Bacillus subtilis HN 503 ,from Xanthomonas campestuis pv. Campestris and from Aspergillus oryzae HN 504 were cloned.Mutator whose alpha-amylase activity and thermostability can be greatly improved was obtained by error-prone PCR. We try to clone alpha-amylase (amyA) gene from Xanthomonas campestuis into Pichia pastoris GS115 to express and secret. Recombinant GSXA was cultured in flask, and the enzyme activity of supertanant reach 33.70u/ml.In this reseach,we first apply LBSP (LB medium with trypan blue) to clone amyA gene and to screen recombinants. An alpha-amylase gene with high enzyme activity was obtained successfully by replacing dNTP with BrdUTP during PCR.
