In the theory of differentitiation of traditional chinese medicine, Tripterygium is the drug of choice for the treatment of rheumatoid arthritis, which is a kind of autoimmune disease. Triptolide is recognized as a principal ingredient responsible for the biological activities of Tripterygium Wilfordii. Howerer, triptolide has strong inhibition in the reproductive system. And steroid hormones play a powerful function in inflammation and reproductive systems. Present project takes triptolide-the main active and toxic ingredient of tripterygium as the research object and uses the transformation of steroid hormone as the leading clue. After establish the model of rheumatoid arthritis in rats, we will observe the exposure, efficacy and toxicity of triptolide and the transformation in the metabolic profiling of steroid hormone on the pathological or physiological state. Then we will illustrate the relationships among them through the further study of molecular mechanisms on these changes. Adopted the technology of LC-MS , GC-MS and chip technology, combined the subjects with pharmacology, toxicology, pharmaceutical analysis and molecular biology and followed the principle of holism in the differentitiation of traditional chinese medicine, we will try to explore the important role of steroid hormone in the efficacy and reproductive of triptolide on the whole and molecular level. The experimental data will use to elucidate the essence and mechanism of the efficacy and reproductive of triptolide which will cast light on the proper use of triptolide for clinical. Under the guidance of traditional chinese theory, the study of tripterygium as a example will be used to reveal the mechanism and regularity of correlations between toxicity and activity of toxic herbs, to establish a safety evaluation model of toxic herbs in line with the international norms and to provide scientific basis for the control of toxicity and reasonable applications of toxic herbs.
类风湿性关节炎是自身免疫性疾病,雷公藤是中医辨证论治该病的首选中药,但对生殖系统具有很强抑制作用,而类固醇激素在炎症和生殖系统中发挥着强大功能。本研究以雷公藤主要活性毒性成分雷公藤甲素为研究对象,以类固醇激素变化为研究主线,建立大鼠类风湿性关节炎模型,考察病理/生理状态下雷公藤甲素暴露量、药效、生殖毒性、类固醇激素代谢轮廓的变化,深入研究之间的关系和主要的分子机制。采用色谱-质谱联用技术和表达谱芯片技术,联合药理学、毒理学、药物分析学、分子生物学等多学科,遵循中医"整体观"原则,从整体水平和细胞分子水平深入探讨类固醇激素在雷公藤甲素和类风湿性关节炎、雷公藤甲素药效和生殖毒中的重要作用。阐明雷公藤甲素抗炎免疫作用和生殖毒的本质及机理,为临床合理应用雷公藤甲素提供实验数据。以中医理论为指导,雷公藤为范例揭示有毒中药毒效相关的作用原理和应用规律,为中医中药控制有毒中药毒性而合理应用提供科学数据。
类风湿性关节炎是自身免疫性疾病,雷公藤是中医辨证论治该病的首选中药,但对生殖系统具有很强抑制作用,而类固醇激素在炎症和生殖系统中发挥着强大功能。本研究以雷公藤主要活性毒性成分雷公藤甲素为研究对象,以类固醇激素变化为研究主线,建立了稳定重现的LC-MS/MS测定血浆和组织中雷公藤甲素和类固醇激素(孕酮、睾酮、皮质酮、皮质醇等)含量的检测方法,采用完全佐剂法建立大鼠类风湿性关节炎模型,选择多个剂量,从药效剂量到毒性剂量(50、100、300、450、600μg/kg)考察病理/生理状态下雷公藤甲素暴露量(体内浓度)、药效(TNF-α、IL-1β和IL-6)、生殖毒性(病理切片、血生化指标)、主要相关类固醇激素代谢轮廓(体内浓度)的变化。研究发现雷公藤甲素对大鼠类风湿关节炎治疗作用明显;激素变化:在造模第1天,孕酮显著增加,之后逐渐下降;第15天时,皮质醇显著增加。给予雷公藤甲素治疗后,在低剂量组,激素变化不明显,而随着剂量的增加,激素水平逐渐下降,第35天时,皮质酮的变化呈现显著差异。随着剂量的增加,毒性尤其是生殖毒显著表现,并且在关节炎大鼠中毒性表现强于正常大鼠。其在类风湿关节炎大鼠中的药代动力学也发生显著改变,吸收增加,但是消除减慢,这一现象与药效和毒性的表现是一致的。通过芯片和分子生物学实验,研究发现采用高剂量给予类风湿关节炎大鼠雷公藤甲素时,相关转运蛋白Oatp1a1、Oatp1b2、Oct1的mRNA水平明显上调。本项目采用多种技术,联合药理学、毒理学、药物分析学、分子生物学等多学科,遵循中医“整体观”理论,深入探讨类固醇激素在雷公藤甲素和类风湿性关节炎、雷公藤甲素药效和生殖毒中的重要作用。目前共已发表文章5篇,其中2篇SCI文章,3篇中文期刊论文。仍有两篇文章正在撰写中。共培养2名硕士研究生。在本研究的基础上课题负责人申请的国家自然科学基金面上项目于2016年获得资助。本项目以中医理论为指导,以雷公藤为范例揭示有毒中药毒效相关的作用原理和应用规律,为临床合理应用雷公藤甲素提供实验数据,为中医中药控制有毒中药毒性而合理应用提供依据。
