No studies regarding molecular phylogeny among the ranid species were reported during the time the present project was approved. The phylogenetic relationships among 24 species of Chinese ranid frogs, 7 species of brown frogs of the genus Rana, and among the haplotypes of Pelophylax nigromaculata inhabiting the middle and lower reaches of the Yangtze River, were studied during the past three years. The NJ tree based on the sequences of 12S rRNA gene fragments indicates that the 24 frogs were divided into three lineages, and supports the establishment of ranid genera Fejervarya, Limnonectes, Hoplobatrachus, Paa, Odorrana, Rugosa and Pelophylax. Therefore, provide molecular edidence supports the taxonomical changes of some groups of traditional Rnna suggested by Fei et al (1990) and Dubois (1992). The NJ and MP trees reconstructed based on sequences of cyt b and 12S rRNA gene fragments indicate that the genus Rana is a monophyletic group, R. chensinensis is most closely related to R. amurensis, and support to place R. zhenhaiensis, R. omeimontis, R. chaochiaoensis and R. longicrus into R. japonica species group. According to the partial sequences of 16S rDNA of Pelophylax nigromaculata, samples from Jiangsu, Henan, Anhui and Zhejiang Provinces share a common haplotype, those from Hubei and Hunan belong to another hpalotype. The Japanese population belongs to the third haplotype.
采用PCR直接测序法测定我国蛙科40余种12S和16SrRNA基因,cytb基因的序列,结合GenBank上有的蛙科物种的相应序列重建蛙科分子系统发生树,在DNA分子水平上讨论蛙科的系统发生关系,对重新划分蛙属的各种意见进行讨论,为蛙科种上分类提供分子证据。研究结果将填补世界上这方面的研究空白。
