The disorder of endocrine-immunomodulation net (EIMN) is closely.related with the whole course of systemic lupus erythmatosus (SLE)..Treating SLE with TCM has an advantage as a whole, and that may be.due to the effect of TCM on EIMN. Therefore, the effect of.langchuangding (LCD) on glucocorticoid (GC) and sex.immunomodulation circuit of SLE was studied. In clinical research, the.patients were randomly divided into 2 groups, one being treated with.LCD plus GC, the other with GC only; to measure the level of.corticosteroid (CS), adrenocorticotropic hormone (ACTH), estradiol (E2),.testosterone (T), prolactin (PRL) by radioimmunoassay; to test.interleukin-1 (IL-1), soluble interleukin-2 receptor (sIL-2R), neopterin,.soluble intercellular adhesion molecule-1 (sICAM-1) by ELISA; to test.periphery blood T lymphocyte subsets and Fas, Bcl-2 by cytoflurography..Meanwhile, MRL/lpr mice were used in experimental study, the level of.estradiol receptor (E2R), glucocorticoid receptor (GCR) in renal tissue.was tested with immunohistochemical technique. The result showed: the.level of CS, ACTH, and T were lower; E2, PRL, sICAM-1 were higher..There were close relationship between the SLAM and the level of E2,.PRL, sICAM-1; and the ratio of CD4.+/CD8.+ was inverted (<1); the level.of Fas of CD3.+ , CD4.+, CD8.+ T lymphocyte were higher abnormally, the.level of Bcl-2 of CD8.+ T lymphocyte was lower obviously. After being.treated with LCD plus GC, the level of CS, ACTH and T could be raised.significantly; PRL, IL-1, sIL-2R, neopterin, sICAM-1 could be reduced..The effect above was more obvious than that of GC group. And the level.of Bcl-2 of CD3.+ , CD4.+ cell was raised obviously, Fas of CD8.+ cell and all.subsets reduced significantly. There were obviously different between the.two groups about the level of CD4.+ and CD8.+, Fas level of CD8.+ cell,.Bcl-2 level of CD4.+ and CD8.+ cell. All of above showed: LCD could.effectively regulate the level of the GC, sex hormones and their receptor,.cytokine, T lymphocyte subsets and Fas, Bcl-2, and had better regulative.effect on EIRN. The regulative effect of TCM was more obvious than that.of GC on the expressive level of GCR and E2R in renal tissue. At the.same time, it was discovered that TCM could effectively improve the.pathogenic state of concentration, viscosity, coagulation and coacervation.in serum. That could be beneficial for TCM to regulate the function of.nervous system, endocrinium and immune system, to reduce the deposit.of immune complex, and to promote the situation of SLE to be stable and.take a turn for the better.

本课题对系统性红斑狼疮(SLE)患者和用诱导法建立动物模型,以ACTH,GC,GCR和IL-1,IL-2,sIL-2R,sICAM-1,apotosis,及Fas-FasL作为主要测定指标,结合形态学变化,观察中药狼疮定冲剂对下丘脑-垂体-肾上腺轴和免疫系统的影响.旨在从糖皮质激素免疫调节环路的角度,探讨狼疮定对SLE的作用机理.
