The balance of Th cells or Th-type cytokines plays an important role in the pathogenesis of systemic lupus erythematosus(SLE).Th cells are regulted by many moleculars through many steps, which also attend the development of autoimmune disease. There was a few reports about that and most of them were not systemic studies. We study the abnormals of the balance of Th cells and regulatory moleculars in the pathogenesis of SLE. Result: We have found that Th1 cell and IFN-γmRNA expression decreased significantly in recent-onset SLE patients. It's maybe attribute to IL-12 and IL-12Rβ2 and IL-18 mRNA expression decreased or abnormal of STAT4, GATA3. Different clinical manifestation maybe exsit different abnormal of Th regulatory moleculars. IL-4R and GATA3 could be used as the index to evalute disease activity in SLE patients.
以红斑狼疮作为研究的对象,研究外周血白细胞IL-12受体β2链的表达及单个细胞水平细胞因子的产生,并对IL-12受体β2基因调控序列多态性进行分析,以探讨调控Th细胞分化的分子基础,并验证IL-12受体β2链能否作为鉴别Th1、Th2的表面标志,为存在Th1、Th2失衡的多种疾病的临床监测及免疫干预治疗提供理论依据。
