Atherosclerosis (AS) cardiovascular disease caused by Hyperlipidemia has become the first killer which being a serious harm to the human health, prevention of the disease has very important significance. Now Dyslipidemia has been identified as the "major independent risk factors" of AS attack, and reverse cholesterol transport (RCT) obstacle is a key link that lead to dyslipidemia and lipid in blood vessel walls, peroxisome proliferation-activated receptor(PPAR) signal pathway plays an important regulatory role in RCT. The new study has found in our team that the Herbal-cake-separated moxibustion could activate the expression of PPARγ protein in rabbit aortic endothelial cells to have the effect of delay the AS plaque formation. In this subject, we clasp the two key links, namely cholesterol outflow in Peripheral cells and selective cholesterol fat intake in Liver cells in RCT, adopting the Herb-partition moxibustion as intervention mean and AS rabbit as study subjects, using many kinds of biological technology such as cell culture, flow cytometry and Western Blot, Real-time fluorescent quantitative polymerase chain reaction and so on, from the signal pathway of PPARγ-LXRα-ABCA1 to study the resistance to atherosclerosis formation mechanism in promoting reverse cholesterol transport with Herb- partition moxibustion, and to provide the new strategy for the research in the prevention thought and the AS pathogenesis.
高脂血症导致的动脉粥样硬化(AS)心血管疾病巳成为严重危害人类健康的第一位杀手,防治本类疾病具有非常重要的意义。血脂异常已被确定为AS发病的"主要的独立危险因素",而胆固醇逆转运(RCT)障碍是导致血脂异常和血管壁脂质沉积的关键环节,过氧化酶体增殖物激活型受体(PPAR)信号途径又在RCT中起着重要的调控作用。课题组最新研究发现,隔药饼灸能激活AS兔主动脉内皮细胞中PPARγ蛋白表达从而起到延迟AS斑块形成的作用。本项目紧扣RCT过程中的两个关键环节,即外周细胞胆固醇外流与肝细胞胆固醇脂的选择性摄取,采用隔药饼灸,以AS兔为受试对象,运用细胞培养、流式细胞术、蛋白质印迹、实时荧光定量聚合酶链反应等多种生物技术手段,从PPARγ-LXRα-ABCA1信号途径研究隔药饼灸促胆固醇逆转运抗动脉粥样硬化形成机制,为AS发病机制与防治思路的研究提供新的策略。
