Rheumatoid arthritis (RA) is a chronic teratogenic autoimmune disease characterized by articular synovial hyperplasia leading to synovial inflammation as a pathological feature, recent studies suggesting that its onset is closely related to the secretion of inflammatory factors by synovial fibroblasts (FLS) and autophagy and Abnormal apoptosis of FLS cells. Sanbaibang(Toddalia asiatica) is effective in Clinical treatment for RA .Our preliminary study found that alcohol extraction of Sanbaibang could inhibit the secretion of RA-FLS inflammatory factors and the project team has obtained the technology through the extraction and separation of the active ingredients in Sanbaibang. Therefore, under the guidance of Tujia medicine theory, the hypothesis of science that Active ingredients of Sanbaibang can enhance the Autophagy and reduce proliferation in Rheumatoid Arthritis Fibroblast-like Synoviocytes by PI3K/Akt/mTOR pathway is put forward.Combinating with pharmacology, molecular biology and other multidisciplinary research methods, advanced techniques such as plasmid transfection and flow cytometry will be used to deeply study the mechanism about active ingredients of Sanbaibang regulating the Autophagy and Apoptosis in Rheumatoid Arthritis Fibroblast-like Synoviocytes by PI3K/Akt/mTOR pathway which will provide scientific basis for the further development use of Tujia medicine .
类风湿性关节炎(RA)是一种以关节滑膜增生导致滑膜炎症为病理特征的慢性致畸性自身免疫性疾病,近年来研究认为其发病与滑膜成纤维细胞(FLS)分泌炎性因子,以及FLS细胞的自噬和凋亡异常有密切关系。土家族药物三百棒临床用以治疗RA疗效确切。项目组前期研究发现三百棒醇提物能抑制RA-FLS炎性因子的分泌,并通过提取分离获得三百棒有效成分群。因此本课题在土家族医药理论指导下,提出“三百棒有效成分群可以通过PI3K/Akt/mTOR信号通路增强RA-FLS的自噬,降低炎性因子的分泌,促进其凋亡,从而起到治疗RA滑膜炎症作用”的科学假说。拟结合药理学、分子生物学等多学科研究手段,采用质粒转染、流式细胞术等先进技术,从细胞、整体动物及分子不同水平,深入探讨三百棒有效成分群通过调控FLS自噬与凋亡水平以治疗RA的分子作用机制,为进一步开发和利用土家医药提供科学依据。
