Rheumatoid arthritis (RA) is a chronic teratogenicity of multiple autoimmune diseases. The clinical curative effect of compound Sanbaibang (Toddalia asiatica)medicinal liquor and Jinbianqufengyin for the treatment of RA is exact, but the mechanism study of Sanbaibang in the treatment of RA is still not deep enough. under the guidance of Tujia medicine theories, combined with previous research results, The project team put forward a scientific hypothesis of " To achieve the treatment effect of RA, Sanbaibang inhibited the stimulation of inflammatory midiators on synovial fibroblats(FLS),reduce the abnormal proliferation of synovial membrane, through regulating the expression of miRNA-146a and negative regulating the TLR/NF-κB pathway ". So this research method will combine with multi-disciplinary research of Tujia medicine, molecular biology, pharmacology, from three levels including the whole animal model, cell, molecule, We will use plasmid transfection, flow cytometry and other advanced technologies, reveal Sanbaibang on the expression of TLR/NF-κB signaling pathway and the effect of miRNA146a in synovial cells of RA, elucidate the molecular mechanism in the treatment of RA, and provide scientific basis for the clinical application of Tujia medicine for the treatment of RA.
类风湿性关节炎(RA)是一种多发的慢性致畸性自身免疫性疾病,临床以关节滑膜炎为病理特征。以土家族药物三百棒为主药的复方三百棒药酒、金边祛风饮治疗RA临床效果确切,但目前对三百棒治疗RA的机制研究尚不够深入。项目组在土家族医药理论指导下,结合前期研究成果,提出"三百棒可以通过调节miRNA-146a的表达,负调控TLR/NF-κB信号通路,以抑制炎性介质对滑膜成纤维细胞(FLS)的刺激,减少滑膜异常增殖,达到治疗RA作用"的科学假说。故本课题结合土家医学、分子生物学、药理学等多学科研究手段,从整体动物模型、细胞、分子三个层次,采用质粒转染、流式细胞术等先进技术,揭示三百棒对RA中滑膜细胞的TLR/NF-κB炎症信号通路及miRNA146a表达的影响,以期阐明三百棒治疗RA的分子作用机制,为土家医药临床治疗RA提供科学依据。
类风湿性关节炎(RA)是一种多发的慢性致畸性自身免疫性疾病,临床以关节滑膜炎为病理特征。以土家族药物三百棒为主药的复方三百棒药酒、金边祛风饮治疗RA临床效果确切,但目前对三百棒治疗RA的机制研究尚不够深入。项目组在临床研究中通过实时荧光定量PCR及基因芯片等方法,发现RA的发病与患者外周血中miR-146a和miR-155的表达上调,以及差异表达的lncRNA有关,但与miR-146a单核苷酸多态性无关。在动物实验中通过急毒实验发现三百棒口服急性毒性较低;通过对RA动物模型口服和外敷三百棒提取物,发现三百棒能通过抑制IL-1、TNF-α、IL-8等促炎因子的表达,抑制VEGF、MMP-3的表达,调节其机体外周血中T淋巴细胞表面特异性抗原,从而抑制RA滑膜细胞中成纤维细胞的异常增殖,并促进其凋亡。在成分研究中通过活性成分分离鉴定,发现三百棒治疗RA的有效成分主要是生物碱类,如白屈菜红碱。在细胞实验中通过用脂多糖刺激MH7A细胞和THP1细胞制作RA滑膜炎性细胞模型,将三百棒醇提液共培养后发现三百棒确实能提高miRNA-146a的表达,抑制TLR/NF-κB信号通路中TLR2、TLR4、MyD88、IRAK-1、TNAF-6、NF-κB、IL-6、MMP-9等相关因子的表达,促进RA滑膜细胞中成纤维细胞凋亡,抑制其异常增殖,改善RA关节滑膜炎的症状。这可能是三百棒治疗RA的作用机制之一。
