Using the methods of RACE, real time PCR ,RNA interference and so on,this research is intended to further study the interaction between the lipopolysaccharide and beta-1, 3 - glucan binding protein (LGBP) and integrin in the Chinese mitten crab,Eriocheir sinensis,based on our previous work;and on this basis, we want to explore the possible immune signal transduction pathway;at the same time, in order to elucidate the immune recognition molecular mechanism of LGBP,we will describe the whole process that how LGBP is interacted with integrin and how the immune signal is transducted,which ultimately leads to immune effector genes playing a role in innate immune response;this research will provide a theoretical basis to find a scientific disease prevention and control measure,improve resistance to diseases, and achieve the sustainable development of the aquaculture industry; this study has a very important theoretical significance to in-depth understanding of the innate immune function and regulation mechanism in invertebrates and improving the crustacean immunology content.
本课题拟在申请人前期工作基础上,采用RACE、实时荧光定量PCR和RNA干扰等生物技术,进一步研究中华绒螯蟹脂多糖和β-1,3-葡聚糖结合蛋白(LGBP)与整合素之间的相互作用关系,同时分析由此作用引起的机体抗氧化酶系统的变化规律;并探讨此过程中免疫信号的传递途径;结合中华绒螯蟹中已鉴别的免疫效应相关基因,通过研究LGBP-整合素-免疫效应基因的相互作用关系,阐明LGBP从识别外源病原后与整合素相互作用,进行信号传递,并最终引起免疫效应基因发挥作用的整个过程,从而系统阐述中华绒螯蟹免疫识别作用的分子机制;为寻找更科学的蟹病防治措施、提高养殖对象抗病能力,实现养殖产业的可持续发展提供理论基础;本研究对深入了解无脊椎动物的先天性免疫功能与调节机制和完善发展甲壳动物免疫学内容都具有十分重要的理论意义。
中华绒螯蟹( Eriocheir sinensis ) 作为我国重要增养殖对象之一,近年来,关于其免疫机制的研究越来越受到人们的重视。本项目采用SSH、RACE、实时荧光定量PCR等生物技术手段,构建了嗜水气单胞菌感染前后中华绒螯蟹抑制削减杂交文库和转录组文库并进行了生物信息学分析,筛选获得本研究所需的免疫识别蛋白LGBP基因、整合素基因,免疫信号传递途径相关Dorsal和Tube基因及免疫效应相关PPAF、PXN和SOD、GPX、GST基因阳性克隆,并进一步克隆获得上述基因cDNA全长和时空表达变化规律,研究结果表明,LGBP从识别外源病原后与整合素相互作用,进行信号传递,并最终引起免疫效应基因发挥作用的整个过程。本研究结果较为系统的阐述了中华绒螯蟹免疫识别作用的分子机制,为寻找更科学的蟹病防治措施、提高养殖对象抗病能力,实现养殖产业的可持续发展提供理论基础;本研究结果也对深入了解无脊椎动物的先天性免疫功能与调节机制和完善发展甲壳动物免疫学内容都具有十分重要的理论意义。
