As an excellent substitute of metal implantable materials, , however, the biomechanical property and osseointegration of polyetheretherketone (PEEK)-based composite should be further enhanced. Therefore, to prepare the PEEK-based composite with high osteogenic activity, our group plan to employ bone forming peptide-1 (BFP-1) and dexamethasone liposomes (Dex/Lip) to modify carbon fiber-reinforced polyetheretherketone–nano hydroxyapatite (PEEK/n-HA/CF), which is a ternary composite having good biomechanical property and osteogenic activity, via dopamine chemistry. X-ray photoelectron spectroscopy (XPS) will be utilized to identify the chemical constituent and elemental state of BFP-1 and Dex/Lip. Simultaneously, human bone marrow mesenchymal stem cells will serve as the cell model, and real-time PCR and western blotting will be used to detect the expression of bone-related gene and proteins of cells, respectively. The in vivo osteogenesis ability of the decorated PEEK/n-HA/CF biocomposite will be probed through Micro CT and immunohistochemical approaches. The synergistic effect of BFP-1 and Dex/Lip will be computed by the dual factorial analysis. These had paved a novel way to develop bone repair materials and present a new idea to treat the bone disease in clinic.
作为替代金属类骨植入材料的高分子材料,聚醚醚酮基复合材料的生物力学性能和骨整合效果仍有待进一步提高。在获得具有良好生物力学性能和一定成骨活性的碳纤维增强聚醚醚酮/纳米磷灰石三元复合材料的基础上,课题组拟利用成骨分化小分子物质--成骨多肽BFP-1和地塞米松脂质体,通过聚多巴胺实现其在材料表面的化学接枝,以期建立成骨分化小分子修饰材料表面的技术,获得骨整合效果优异的聚醚醚酮三元复合材料植入物。课题将通过XPS等对地塞米松脂质体及成骨多肽的接枝情况进行表征;以人骨髓间充质干细胞为模型细胞,采用RT-PCR及Western Blotting等检测细胞相关成骨基因和蛋白表达,茜红素染色检测钙结节,并通过二元析因统计分析BFP-1和地塞米松是否具有协同作用,探讨其分子机制;利用比格犬颌骨植入模型,采用Micro CT、免疫组化等验证修饰后植入物的骨整合效果,为拓展聚醚醚酮复合材料的临床应用奠定基础。
