Recombinant yeast cell transformed respectively with plasmids encoding the human estrogen receptor, androgen receptor, progestogen receptor and thyroxine receptor and its corresponding hormone response element linked to a reporter gene were constructed and evaluated for screening the hormone-like activities of environmental endocrine disruptors(EEDs) mediated by hormone receptors. Based on measuring the expressed product of reporter gene, the recombinant yeast system proves preliminarily a promising assay for rapid and easy screening of widely existing EEDs and also for correlative mechanistic studies.
本研究拟采用分子生物学方法制备四种分别含有雌激素、雄激素、孕激素、甲状腺素受体表达质粒和其相应的激素作用反应质粒的重组酵母细胞株,建立可简便、高效、快速地检测具有干扰上述四种激素作用的环境内分泌干扰物的重组酵母检测系统。该系统可用于筛检环境内分泌干扰物,研究其作用机制,对促进毒理学的发展具有重要的理论和实际意义。
