Based on the preliminary experimental results of the two projects (NNSF: 81160250 & Thousands of Foreign Experts Plan: WQ20122200065), we found that the human lung cancer specimens exhibited a robust expression of ZFP91 compared with adjacent tissues; Furthermore, ZFP91 induces HIF-1α activation through ZFP91 interacts with TRAF6 and promotes the K63-linked ubiquitination of HIF-1α; Moreover, ZFP91 induces noncanonical NF-κB activation through ZFP91 interacts with HIF-1α and promotes the K48-linked ubiquitination of TRAF3, so it is believed that the cross-talk between NF-κB and HIF-1α signaling pathway plays an important role in development of lung cancer. However, its exact mechanism remains unknown. In this study, we will use the lung cancer tissue microarray, gene overexpression, RNA interference and immunoprecipitation to determine expression of ZFP91 and its clinical significance in lung cancer, and to determine how ZFP91 mediates cross-talk of NF-κB-HIF-1α pathways to regulate lung cancer development, using xenograft tumor models to explore the role of ZFP91 and its immune complexes in lung cancer growth and metastasis. We will also explore the intervention mechanism of chelidonine. This research may extend our understanding on the molecular mechanisms underlying the diverse biological activities of ZFP91-NF-κB-HIF-1α pathways in lung cancer microenvironment.
基于前期国家自然基金(81160250)和外专千人计划(WQ20122200065)研究中重要发现与不足:发现肺癌组织中ZFP91高表达;ZFP91-TRAF6作用后使HIF-1α泛素化(K63)活化,ZFP91-HIF-1α作用后使TRAF3泛素化(K48)降解,因此认为ZFP91介导NF-κB-HIF-1α信号通路交叉对话在肺癌发生发展中起重要的作用,然而其确切机理尚不明确。本研究拟利用肺癌组织芯片、基因过表达、RNA干扰和免疫共沉淀等分析技术,以ZFP91及其免疫复合物TRAF3、TRAF6和HIF-1α为切入点,分析其与临床病理的相关性,研究ZFP91介导NF-κB-HIF-1α信号通路交叉对话在肺癌发生发展中作用,并利用裸鼠移植瘤模型探讨ZFP91及其免疫复合物在肺癌生长中的作用,同时探索白屈菜碱干预机制,以期为ZFP91-NF-κB-HIF-1α通路为靶点的肺癌诊治提供新策略。
白介素-1β(IL-1β)是炎症、感染和自身免疫疾病中具有重要作用的细胞因子。据报道锌指蛋白91(ZFP91)参与各种生物学过程,但对于ZFP91调节IL-1β的表达机制尚未明确,白屈菜碱是我们课题组从长白山药用植物白屈菜根部中提取的一种活性单体化合物,尚未见国内外有解析该种化合物在NF-κB信号转导途径及炎症和肿瘤反应中作用机理的研究报道。本研究(1)利用免疫共沉淀明确观察到外源性和内源性ZFP91、TRAF6、cIAP1和cIAP2的相互作用;利用Western blot法,明确观察到ZFP91能够上调LPS刺激的THP-1细胞和BMDM细胞中pro-IL-1β的产生和活化;明确观察ZFP91促进非经典半胱天冬酶-8(caspase-8)炎症小体成分的表达,并促进在LPS刺激的巨噬细胞中非经典caspase-8炎症小体的集聚;明确观察ZFP91促进LPS刺激的THP-1细胞和BMDM细胞中胞外信号调节激酶-1/2(ERK1/2)、p38促分裂原活化蛋白激酶(p38 MAPK)和c-Jun N-末端激酶(JNK)的活化;利用小鼠模型,明确观察到ZFP91缺失可明显抑制了小鼠炎症表现。其次对(2)在TNF-α诱导状态下,明确观察到白屈菜碱对NF-κB的转录活性有明显的抑制作用;利用Western blot法,明确观察到白屈菜碱可剂量性抑制NF-κB相关基因IκBα、MAPKs、c-IAP1,FLIP、BCL2、ICAM-1、COX-2、MMP-9和Cyclin D1蛋白质表达水平的影响;利用RT-PCR明确观察到白屈菜碱抑制iNOS,IL-8和MCP1 mRNA的表达;利用免疫荧光实验,明确观察到白屈菜碱可抑制TNF-α诱导的p65的核易位;利用流式细胞仪分析技术,明确观察到白屈菜碱可促进TNF-α诱导细胞凋亡;利用平板克隆形成试验和Edu实验,明确观察到白屈菜碱可抑制细胞增殖;(3)本研究团队在进行白屈菜碱抗炎、抗肿瘤筛选中发现一些其他类型的单体化合物,如Mollugin、Shikonin、Amorfrutin A、Dictamnine、Fraxinellone、Convallatoxin、Panaxadiol、Curcumol和Erianin等有较好药理活性,揭示了其作用机理,为后续的抗炎、抗肿瘤药物的开发提供了理论基础。
