Atherosclerosis (AS) is a kind of chronic immune inflammatory cardiovascular diseases which is the main cause of high morbidity of cardiovascular disease and mortality. Current researches have confirmed that Caveolin-1 induced-deacetylation is related to the protective effects on AS via improving endothelial function, inhibiting endothelial cell activation, stabling plaques, decreasing cholesterol intake and foam cell formation, inhibiting expressions of endothelial tissue factor and proinflammatory factor. TiaopiHuxin decoction indicating that it has significant benefit in anti-AS. However, there is no study about the effect of this formula on AS via Caveolin-1 regulatory pathway has not been reported. Based on the important cardiovascular protective effects of Caveolin-1 and its target genes NF-κB、eNOS、LXR, the present study raised hypothesis that TiaopiHuxin decoction has protective effects on AS via deacetylation induced by up regulation of Caveolin-1 gene expression in result to regulation of downstream proteins. The results of this study is expected to clarify that Caveolin-1 is one of the initiating factors of AS and to provide new targets and ideas for AS treatment of traditional Chinese medicine.
动脉粥样硬化(AS)是一种多原因的慢性免疫炎症性心血管疾病,是引起心血管疾病高发病率与死亡率的主要原因。目前研究已证实Caveolin-1在改善内皮功能、抑制内皮细胞活化、稳定斑块、降低胆固醇摄取和泡沫细胞形成、抑制促炎因子表达等多方面发挥其对AS的保护作用。前期承担的课题结果提示中药调脾护心方针对冠心病疗效理想,并在抑制炎症通路方面做了部分试探性研究。目前,此方通过调控Caveolin-1改善AS的作用尚未见报道。基于NF-κB、eNOS、LXR等为Caveolin-1靶基因,故推测Caveolin-1具有重要的心血管保护作用,本项目综合国内外研究提出“调脾护心方通过干预Caveolin-1基因表达调控一系列下游蛋白是其抗AS的可能作用机制”。通过对Caveolin-1的深入研究有望阐明Caveolin-1是AS发病的始动因素,并为中药治疗AS的研究提供新的靶点和思路。
