Based on the fact that Traditional Chinese Medicine meridian theory that Qiao Channel controls sleep, which play an important role in controlling sleep-rhythm and purinergic signalling molecules adenosine and adenosine receptors are recognized as key sleep regulators, we proposed a hypothesis that "adenosine-adenosine receptors would be the neurobiological basis for Qiao Channel controls sleeping by regulating sleep rhythm". Focusing on this hypothesis and guided by Qiao Channel theroy, we will curry out acupuncture on Shenmai and Zhaohai acupoints of Qiao Channel on insomnia rat model, and then apply modern tecnologies such as Chronobiology, molecular biology,and neurobiology,agonist and antagonist,etc., to observe the effect of Qiao Channel acupuncture on sleep-wake cycle, and to reveal the role of adenosine and adenosine receptors and SCN circadian regulation in sleep regulation induced by Qiao meridian-acupuncture ,which maybe could provide clues to the neurobiological basis mechanism. the modern neurobiological basis of the theory of Qiao Channel controls sleeping.
传统经络理论认为,跷脉司寤寐,对睡眠节律的调整是其重要的临床价值。腺苷及其腺苷受体是公认的睡眠调节因子。基于此,我们提出“腺苷-腺苷受体是介导跷脉司寤寐调整睡眠节律神经生物学基础”工作假说。围绕该假说,我们将以跷脉司寤寐理论为指导,构建失眠小鼠模型,采用针刺调理跷脉申脉、照海穴位治疗,并应用现代时间生物学、分子生物学、神经生物学、受体激动与阻断、等方法技术,研究跷脉针刺对睡眠-觉醒周期的调节作用,明确跷脉针刺调整睡眠节律腺苷及其腺苷受体的动态变化,揭示是否通过对视交叉上核的节律调整实现腺苷-腺苷受体参与跷脉针刺改善睡眠的作用及其分子神经生物学机制,科学阐释跷脉司寤寐理论的现代神经生物学基础。
