The research about the biological basis of acupuncture is an important point of revealing the principle of acupuncture effect.Acupuncture time-space effect research is a new research model of acupuncture effect.Microdialysis technology is one of the most accurate technical means of testing the central neurotransmitter by positioning and quantitative analysis.So our project focused on the research point about the change of the central neurotransmitter's content on focal cerebral ischemia by acupuncture treating.Using the techology of microdialysis,observing and analyzing the concentration of lactic acid,pyruvic acid and the content of Glu,GABA on the side and the other side of corpus striatum of focal cerebral ischemia before and after acupuncture.Proving regulating effect of acupuncture on the central enviroment of focal lesion and sqreading depression in remote encephalic region.Observing and analyzing the content of the central neurotransmitter in focal cerebral ischemia and remote encephalic region at the different time after acupuncture Shuigou (GV 26),drawing a curve chart of time-space effects.Proving there is the rule of acupuncture immediate -additive time effect and space effect on the treating of focal cerebral ischemia.
探讨针灸效应的生物学基础,是揭示针灸疗效原理的重要切入点;针刺时空效应研究是针刺效应规律研究的全新模式。微透析技术是目前最为准确的定位定量分析检测中枢生命活性物质的技术手段之一,故本项目拟以针刺调节局灶性脑缺血的脑内微环境神经递质变化为研究切入点,采用中枢微透析实时采样技术,观察分析针刺对局灶性脑缺血大鼠缺血脑组织及对侧相应脑区脑组织间液乳酸、丙酮酸浓度和谷氨酸、γ-氨基丁酸2种氨基酸含量的变化,验证针刺对局灶性脑缺血局部及扩散性抑制影响的远隔脑区微环境的调节作用;观察分析针刺"水沟"穴后不同时段局灶性缺血脑组织区、缺血诱导的扩散性抑制引起的远隔脑区脑组织细胞外液神经递质含量的动态变化,分别绘制时空效应曲线。
