Laryngeal cancer is a common malignant tumor of the head and neck, especially the mortality of advanced laryngeal cancer is still high, therefore, it is urgent to find a reliable molecular target of laryngeal cancer and to elucidate its molecular mechanism. We have been working on the regulation of non-coding RNA in laryngeal cancer. Previous study confirmed that downregulated-miR-194 dependent upregulation of WEE1 led to the development of laryngeal cancer (J Hematol Oncol, 2017). However, the mechanism of down-regulation of miR-194 remains unclear. CircRNA chip data revealed that circ_0006783 was overexpressed in laryngeal cancer and positively correlated with WEE1. Bioinformatics analyses suggested a possible competitive binding of circ_0006783 to miR-194. Based on the previous research, we intend to clarify the mechanism that circ_0006783 upregulates WEE1 by base pairing with miR-194, thus promoting the malignant progression of laryngeal cancer, in order to provide new theories and therapeutic targets for the treatment of laryngeal cancer.
喉癌是头颈部常见的恶性肿瘤,尤其是晚期喉癌死亡率较高,因此,寻找喉癌可靠的分子靶点及阐明其分子机制迫在眉睫。申请人致力于喉癌非编码RNA调控方面的研究。前期发现低表达miR-194可致WEE1基因表达上调促进喉癌的发生发展(J Hematol Oncol,2017),但miR-194下调的机制仍不明确。circRNA芯片对人喉癌组织进行表达谱筛选时发现circRNA_0006783在喉癌组织中过度表达,且其表达与WEE1基因的表达呈正相关。生物信息学分析提示circ_0006783与miR-194存在竞争性结合位点。本项目拟在前期研究基础上,采用RIP、RNA-FISH、荧光素酶报告基因等实验方法从新的视角揭示circ_0006783通过与miR-194竞争性结合,阻断miR-194对其靶基因WEE1的抑制作用促进喉癌发生发展的分子机制,为逆转喉癌恶性进程提供新思路和新靶点。
头颈鳞癌常见的恶性肿瘤,尤其是晚期喉癌死亡率较高,因此,寻找头颈鳞癌可靠的分子靶点及阐明其分子机制极其重要。CircRNA 作为非编码 RNA 的重要组成部分,在头颈鳞癌中充当了重要的角色。circTNS4(chr17:38652238-38652772)由基因TNS4外显子2 环化形成。RNase R消化实验联合 RT-QPCR 、琼脂糖凝胶电泳证实了circTNS4的环状结构;进一步RT-QPCR检测发现circTNS4在头颈鳞癌中表达下调,且它的表达水平与头颈鳞癌的T分期有关;FISH实验显示 circTNS4 在头颈鳞癌细胞中定位于细胞核和细胞质,circTNS4 的过表达能够抑制头颈鳞癌细胞的增殖、侵袭、迁移, circTNS4的敲低具有相反的效应。另外,circTNS4 的过表达抑制了头颈鳞癌细胞在裸鼠体内的增殖能力,敲低circTNS4则促进头颈鳞癌细胞在裸鼠体内的增殖能力。一系列生物信息学分析发现 miR-5100 是 circTNS4 的靶基因,荧光素酶报告基因实验证实了二者存在结合位点。circTNS4 的低表达上调了头颈鳞癌细胞中miR-5100 的水平,其高表达具有相反的效应。 综述所述,circTNS4可以通过调控miR-5100/SCAI的表达来抑制头颈鳞癌的增殖和迁移,这为头颈鳞癌早期诊断和治疗提供了新的靶点和思路。
