The recent study has reported that circular RNA- circ_0058106 is significantly upregulated in laryngeal cancer tissues. However, its function is still unknown. We found that downregulation of circ_0058106 expression inhibited the invasion and migration of laryngeal cancer cells and also inhibited transforming growth factor-β (TGF-β) induced epithelial mesenchymal transition (EMT). Moreover, we further identified the interaction between circ_0058106 and Twist, a basic helix-loop-helix transcription factor by using RNA pull down and RNA immunoprecipitation assay. Activating Twist upregulates N-cadherin and downregulates E-cadherin through binding with the E-box in the promotor of N-cadherin and E-cadherin, and subsequently induce EMT. Therefore, we hypothesize that circ_0058106 induce EMT through interacting with Twist to promote the invasion and migration of laryngeal cancer. The aim of this study is to demonstrate this hypothesis and further discover a new target and lay the foundation for the therapy of laryngeal cancer metastasis.

喉癌是目前世界范围内最常见的头颈部恶性肿瘤之一。癌细胞转移是最终导致喉癌患者死亡的最主要原因。本研究以环状RNA(CircRNA)这一生命科学热点为研究切入点来阐明CircRNA参与调控喉癌细胞侵袭和转移的机制。本研究结果显示,Hsa-circ_0058106在50例喉癌组织标本中较正常癌旁组织显著上调表达,且其表达量与患者淋巴结转移、肿瘤临床分期呈现出显著正相关关系。通过将慢病毒包装的shRNA感染喉癌细胞,敲减hsa-circ_0058106表达后,喉癌细胞侵袭和转移能力明显降低,其在裸鼠肺中的转移明显被抑制。此外,我们还发现hsa-circ_0058106沉默可以抑制TGF-β诱导的上皮间质转换(EMT)过程。在机制上,我们发现hsa-circ_0058106在细胞质和细胞核中均有表达,一方面细胞质中的hsa-circ_0058106可以与miR-153特异性结合,通过通过竞争性內源RNA(ceRNA)机制调控Snail1;另一方面核内表达的hsa-circ_0058106可以与转录因子Twist1结合并促进后者的核转位。综上所述,本研究证实了hsa-circ_0058106可以通过ceRNA和调控转录因子核转位双重作用机制调控EMT过程,影响喉癌细胞的侵袭和转移。本课题的完成将有助于阐明环状RNA参与调控喉癌侵袭和转移的分子机制,为阻断喉癌细胞侵袭和转移提供新的治疗靶点和理论依据。
