The synthetic small molecules that can irreversibly and selectively modify nucleic acid have attracted considerable attention in chemistry and biology. Photoinduced DNA cleaving agents are especially convenient since they are inactive until activated by light. In this research, three kinds of novel photoinduced DNA cleaving agents were synthesized in solution. Its ability of DNA cleaving was investigated with Supercoiled DNA (pBluescript SK) under UV irradiation. The DNA cleaving activities of these novel agents are influenced dramatically by the structure of the DNA binding moiety and the concentration of the cleaving agents, and the cleaving activity was enhanced by the hairpin polyamide. The present study has provided a useful method for the synthesis of the novel DNA cleaving agents. They will allow us to probe deeper into the biological phenomena of interactions between small molecules and DNA, making the cleaving agents a very useful investigation tool in chemical biology and gene-selection drug.
