Post-operative nausea and vomiting (PONV) and chemotherapy-induced nausea and vomiting (CINV) seriously affect the quality of life and chemotherapy compliance of patients, but there are still many problems of existing marketed anti-emetic drug, i.e. single target, side effects, expensive price, uncertain safety and efficacy. Recent studies have found that activation of TRPV1 channels can inhibit vomiting, which provided a new breakthrough to develop safe and effective anti-emetic drugs..It was found that new natural TRPV1 agonist decumbensine showed useful anti-emetic efficacy and good safety in vivo, without adversely affecting behavior, blood pressure or temperature control in previous studies. The project intends to study dopamine (DA), 5-hydroxytryptamine (5-HT), substance P (SP), NK-1 receptor, c-fos, CCK, CGRP, and MTL using immunohistochemistry, RT-PCR, western blot and etc., and explore the anti-emetic mechanism of decumbensine. We further investigate the influence of decumbensine on the release of substance P and TRPV1 expression of dorsal root ganglion (DRG) neurons. The project will offer new idea for the development of new safe and effective anti-emetic drugs.
术后恶心呕吐和化疗诱发的恶心呕吐严重影响患者的生活质量和对化疗的依从性,而现有上市止吐药物仍存在着作用靶点单一、毒副反应较大、价格昂贵、安全性与疗效尚需观察等问题。近年研究发现激活TRPV1通道可以抑制呕吐反应,为研发安全有效的止吐药物提供了一个新的突破点。.前期研究中筛选出的新型天然TRPV1激动剂decumbensine在体内实验中具有有效的抗呕吐作用和很好的安全性,且未对动物的行为、血压和体温控制产生不利影响。本项目拟运用免疫组化、RT-PCR、Western blot等方法,以多巴胺、5-羟色胺、P物质、NK-1受体、c-fos、胆囊收缩素、降钙素基因相关肽和胃动素为考察指标,探讨decumbensine的抗呕吐机制;并进一步考察decumbensine对背根神经节神经元P物质释放和TRPV1表达的影响作用。本项目的顺利实施将为研制新的安全有效的多靶点抗呕吐药物提供新的思路。
